摘要
目的:检测急性胰腺炎患者血清中微小RNA-340-3p(miR-340-3p)、CXC趋化因子配体-13(CXCL-13)和CXC趋化因子配体-16(CXCL-16)的表达特征,分析其相关性。方法:选择确诊为急性胰腺炎的患者69例作为观察组,选择经体检健康的成人69例作为对照组。留取所有研究对象的血清标本,应用实时荧光定量PCR法(qRT-PCR)检测miR-340-3p的表达,应用酶联免疫吸附实验法(ELISA)检测CXCL-13、CXCL-16的表达。应用生物信息分析预测miR-340-3p和CXCL-13、miR-340-3p和CXCL-16的靶向关系。结果:miR-340-3p在观察组中的表达量明显低于对照组,CXCL-13和CXCL-16在观察组中的表达量明显高于对照组(均P<0.05)。miR-340-3p、CXCL-13和CXCL-16在观察组不同病变分级中的表达差异有统计学意义(均P<0.05)。观察组中miR-340-3p和CXCL-13(r=-0.63,P=0.032)、miR-340-3p和CXCL-16(r=-0.62,P=0.022)均呈负相关性,CXCL-13和CXCL-16(r=0.61,P=0.014)呈正相关性。生物信息分析显示miR-340-3p与CXCL-13、miR-340-3p与CXCL-16具有可能的结合位点。结论:急性胰腺炎患者血清中miR-340-3p低表达、CXCL-13和CXCL-16高表达,miR-340-3p与CXCL-13、miR-340-3p与CXCL-16具有协同负向作用,参与病变的形成和进展。
Objective:To detect the expressions of microRNA-340-3p(miR-340-3p),CXC chemokine ligand-13(CXCL-13)and CXCL-16 in the serum of patients with acute pancreatitis,and to analyze their correlation and clinical significance.Methods:Sixty-nine patients with acute pancreatitis were selected as the observation group.Sixty-nine healthy adults were selected as the control group.The serum specimens of all subjects were collected.Real-time fluorescence quantitative PCR(qRT-PCR)was applied to detect the expression of miR-340-3p,and enzyme-linked immunosorbent assay(ELISA)was applied to detect the expressions of CXCL-13 and CXCL-16.Bioinformatics analysis was applied to predicte the association between miR-340-3p and CXCL-13,miR-340-3p and CXCL-16.Results:The expression of miR-340-3p was significantly lower in the observation group than that in the control group,and the expressions of CXCL-13 and CXCL-16 were significantly higher than those in the control group(all P<0.05).Expressions of miR-340-3p,CXCL-13 and CXCL-16 showed statistically significant differences in different grades of the observation group(all P<0.05).There was a negative correlation between miR-340-3p and CXCL-13(r=-0.63,P=0.032),miR-340-3p and CXCL-16(r=-0.62,P=0.022)in the observation group.There was a positive correlation between CXCL-13 and CXCL-16(r=0.61,P=0.014)in the observation group.MiR-340-3p and CXCL-13,miR-340-3p and CXCL-16 had possible binding sites by bioinformatics analysis.Conclusion:Acute pancreatitis patients have lower expression of miR-340-3p and higher expressions of CXCL-13 and CXCL-16 in serum.MiR-340-3p and CXCL-13,miR-340-3p and CXCL-16 have synergistic negative effects,are involved in the formation and progression of lesions.
