摘要
牛蒡子苷元具有抗炎活性,可以竞争性地抑制弹性蛋白酶活性。通过酶促反应动力学、紫外-可见吸收光谱、圆二色光谱、红外光谱以及分子对接模拟的方法研究了牛蒡子苷元对弹性蛋白酶的抑制作用。结果表明,牛蒡子苷元对弹性蛋白酶是竞争性抑制,K_(i)=6.19×10^(-4) mol/L,IC_(50)=1.25 mmol/L,且两者之间存在π-阳离子、氢键、疏水作用、范德华力多种作用力。
Arctigenin has anti-inflammatory activity,and can competitively inhibit elastase activity.In this paper,the inhibition of arctigenin on elastase was studied by enzymatic reaction kinetics,UV-Vis absorption spectrumum,circular dichroic spectrum and molecular docking simulation.The results showed that arctigenin was a competitive inhibitor of elastase,K_(i)=6.19×10^(-4) mol/L,IC_(50)=1.25 mmol/L,and there wereπ-cations,hydrogen bonds,hydrophobic interaction,electrostatic interaction and Van der Waals between them.
