摘要
根据硼砂-氢氧化钠缓冲溶液中半胱氨酸与碱性三苯甲烷染料孔雀石绿的加成反应,首次提出加成反应褪色分光光度法测定半胱氨酸含量。反应前后体系的最大吸收波长均为616 nm,选择616 nm为测定波长。由于反应生成了无色硫醚衍生物,使得溶液褪色,体系吸光度降低,据此建立分析方法。半胱氨酸在质量浓度2.00~12.0 mg·L^(-1)范围内,与吸光度差值ΔA呈线性关系,相关系数r=0.9993,表观摩尔吸光系数为1.006×10^(4) L/(moL·cm),方法检出限为0.089 mg·L^(-1)。该法用于复方氨基酸注射液、半胱氨酸胶囊中半胱氨酸含量的测定,测定值的相对标准偏差(n=6)均在3%以内,方法快速简便、灵敏度高,结果满意。
Based on the addition reaction of cysteine with the basic triphenylmethane dye malachite green in borax-sodium hydroxide buffer solution,the first addition reaction fading spectrophotometric method was proposed for the determination of cysteine content.The maximum absorption wavelength of the system before and after the reaction is 616 nm,and 616 nm is selected as the measurement wavelength.Colorless thioether derivatives were formed in the reaction,which made the solution fade and the absorbance of the system decreased,so an analytical method was established.Cysteine was linearly correlated with the absorbance difference Δ A at mass concentrations of 2.00~12.0 mg·L^(-1) with a correlation coefficient r =0.9993 and an apparent molar absorbance coefficient of 1.006×10 4 L/(moL·cm),the detection limit of the method is 0.089 mg·L^(-1) .The method was used for the determination of cysteine content in compound amino acid injection and cysteine capsule,and the relative standard deviations( n =6)of the measured values were within 3%.The method is rapid,simple,sensitive and satisfactory.
