摘要
目的:通过LC-MS/MS法探究炮制和煎煮方式对化痰祛湿活血方中20个成分含量的影响。方法:采用LC-MS法建立了测定化痰祛湿活血方中柴胡皂苷a、大黄酚、柴胡皂苷b2、23-乙酰泽泻醇C、柴胡皂苷b1、泽泻醇A、金丝桃苷、槲皮苷、橙皮苷、阿魏酸、橙黄决明素、丹酚酸B、槲皮素、绿原酸、水飞蓟宾、23-乙酰泽泻醇B、丹参酮ⅡA、柴胡皂苷d、山柰酚、芦丁20个成分含量的方法,对比分析将方中泽泻、丹参、郁金、柴胡换成麸炒泽泻、酒丹参、醋郁金、醋柴胡后单煎、混煎4组样品含量的差异。采用Acquity UPLC?BEH RP18色谱柱,流动相为乙腈-0.1%甲酸水梯度洗脱,电喷雾离子源,正负离子模式,分析炮制前后、单煎、混煎的化痰祛湿活血方中20个成分的变化。结果:20个成分在各自浓度范围内线性关系良好,平均加样回收率为95.33%~101.30%。炮制前后、单煎、混煎造成化痰祛湿活血方中20个成分含量有明显差异,其中阿魏酸、柴胡皂苷d、丹酚酸B、水飞蓟宾、泽泻醇A、丹参酮ⅡA、23-乙酰泽泻醇C、槲皮素对组间差异贡献很大。结论:本研究所建立的方法快速、准确、灵敏度高、重复性好,可为化痰祛湿活血方质量标准的建立提供科学依据;同时,炮制前后、单煎、混煎存在明显的差异,炮制对中医临床方剂影响很大。
Objective:To explore the effects of processing and decoction on the contents of 20 components in Huatan Qushi Huoxue Recipe by LC-MS/MS method.Methods:The LC-MS/MS method was used to establish the determination of saikosaponin a,chrysophanol,saikosaponin b2,23-acetylalismol C,saikosaponin b1,alismol A,hyperoside,quercetin,hesperidin,ferulic acid,orange cassiae,salvianolic acid B,quercetin,chlorogenic acid,silibinin,23-acetylalismol B,tanshinone IIA,saikosaponin d,kaempferol,and rutin,to compare and analyze the recipe of Alisma orientalis,salvia,turmeric,and Bupleurum spp.fried with bran,danshen,vinegar,turmeric,vinegar and Bupleurum,and then single decoction.The difference of the sample content of the 4 groups of mixed decoction.Using Acquity UPLC?BEH RP18 chromatographic column,the mobile phase was acetonitrile-0.1%formic acid water gradient elution,electrospray ion source(ESI),positive and negative ion mode,analysis before and after processing,single decoction,mixed decoction of phlegm,removing dampness and promoting blood circulation changes in 20 ingredients.Results:The 20 components had a good linear relationship within their respective concentration ranges,and the average sample recovery rate was 95.33%-101.30%.Before and after processing,single decoction and mixed decoction resulted in significant differences in the content of 20 components in the prescription for removing phlegm,removing dampness and promoting blood circulation.Among them,ferulic acid,saikosaponin d,salvianolic acid B,silybin,alismanol A,tanshinone I A,23-acetylalismol C,and quercetin contributed greatly to the difference between the groups.Conclusion:The method established in this research is fast,accurate,highly sensitive,and reproducible.It can provide a scientific basis for the establishment of the quality standard of Huatan Qushi Huoxue Recipe.At the same time,there are obvious differences between before and after processing,single decoction and mixed decoction.Processing has a great influence on the clinical prescriptions of traditional Chinese medicine.
