姜黄素抗肺纤维化作用的分子机制研究进展 被引量:12

摘要 肺纤维化是一种以成纤维细胞增殖分化、细胞外基质(extracellular matrix,ECM)过量沉积、肺结构及功能受损为主要病理特征的慢性肺间质疾病。虽然吡非尼酮和尼达尼布等抗肺纤维化药物已在临床应用,但仍未达到满意的治疗效果。因此,寻找一种高效低毒的抗纤维化药物一直是临床的迫切需要。姜黄素是从药用植物姜黄Curcuma longa、郁金C.wenyujin及莪术C.phaeocaulis的块茎中提取的一种天然多酚化合物,具有抗炎、抗菌、抗氧化、调血脂、抗癌、抗纤维化等药理活性,研究表明其可通过抑制成纤维细胞增殖分化、调控细胞凋亡、抑制炎症反应、调节转化生长因子-β、抑制氧化应激、抑制ECM沉积等途径有效缓解肺纤维化进展。主要综述了姜黄素抗肺纤维化作用分子机制的研究进展,并讨论了临床治疗肺纤维化的新靶点和思路,以期为姜黄素进一步抗肺纤维化机制的研究及临床应用提供参考。 Pulmonary fibrosis is a chronic lung interstitial disease characterized by fibroblast proliferation and differentiation,excessive deposition of extracellular matrix(ECM),and impaired lung structure and function.Although some anti-fibrotic drugs such as pirfenidone and nintedanib have been used in clinic,they have not achieved satisfactory therapeutic effects.Therefore,there is an increasingly urgent need to develop anti-fibrotic drugs with promising clinical efficacy and favorable safety profile.Curcumin is a natural polyphenolic compound isolated from the medicinal plants Curcuma longa,C.wenyujin,and C.phaeocaulis,and demonstrates significant anti-inflammatory,anti-bacterial,antioxidant,anti-lipid,anti-fibrotic and other pharmacological activities.Curcumin alleviates the progression of pulmonary fibrosis by inhibiting fibroblast proliferation and differentiation,regulating cell apoptosis,inhibiting inflammatory response,regulating transforming growth factors,inhibiting oxidative stress,and inhibiting ECM deposition.Research progress on molecular mechanism of curcumin against pulmonary fibrosis was summarized,and new targets and ideas for treatment of fibrotic diseases in clinic were discussed in this paper,in order to provide reference for further study of mechanism of curcumin against pulmonary fibrosis and its clinical application.
