摘要
目的建立动物源性食品中氯霉素、甲硝唑和氟苯尼考同时检测的液液萃取-液相色谱串联质谱方法,并应用于分析深圳市售禽肉样品中各目标物残留量。方法在试样中加入d5-氯霉素、d4-甲硝唑和d3-氟苯尼考,经液液萃取、净化、浓缩、过滤后,样液注入液相色谱—串联质谱进行检测,内标法定量。结果在优化的实验条件下,方法检出限为0.05μg/kg(S/N=3),定量限为0.1μg/kg(S/N=10),线性范围为0.25~5.0 ng/ml,相对标准偏差小于7%,样品加标回收率在90%~110%之间。结论该方法快速、灵敏、准确,可同时测定禽肉样品中的氯霉素、甲硝唑和氟苯尼考的含量。
Objective To establish a method for simultaneous determination of chloramphenicol,metronidazole and florfenicol in food of animal origin using liquid-liquid extraction coupled with liquid chromatography-tandem mass spectrometry(LC-MS/MS),and to apply this method in analyzing the residues of target substances in poultries sold in Shenzhen.Methods D5-chloramphenicol,d4-metronidazole and d3-florfenicol were added into the sample,respectively.The sample solution was injected into LC-MS/MS for detection after liquid-liquid extraction,purification,concentration and filtration.The internal standard method was used for quantification.Results Under optimized experimental conditions,the limit of detection(S/N=3)of this method was 0.05μg/kg.The limit of quantitation(S/N=10)was 0.1μg/kg.The linear range was from 0.25 to 5.0 ng/ml.The relative standard derivation was less than 7%,and the sample spiked recovery rate ranged from 90%to110%.Conclusions This method is rapid,sensitive and accurate for the simultaneous determination of chloramphenicol,metronidazole and florfenicol in poultries.
