摘要
目的 探讨葛根复方对实验性自身免疫性重症肌无力(experimental autoimmune myasthenia gravis, EAMG)大鼠骨骼肌及线粒体动力相关蛋白1(dynamin-related protein 1, Drp1)的影响。方法 将大鼠随机分为空白组、模型组、葛芪组、葛根组、葛参组、葛芪参妙组、葛妙组、强的松组,共8组,每组各10只。除空白组外,其余各组采用人工鼠源性AchR-α亚基97-116肽段乳化剂进行EAMG模型构建。每天给予葛芪组、葛根组、葛参组、葛芪参妙组、葛妙组、强的松组相应的药物灌胃,空白组和模型组给予生理盐水灌胃,每日1次,连续28天。治疗结束后,采用苏木精—伊红染色(hematoxylin-eosin staining, HE)染色法对大鼠骨骼肌组织进行染色观察、透射电镜观察线粒体超微结构以及蛋白免疫印迹(Western blot, WB)法检测各组大鼠线粒体Drp1蛋白的表达。结果 与模型组比较,葛根各组及强的松组骨骼肌组织排列紊乱、核偏移、炎症浸润减少,线粒体肿胀变形、嵴结构不清、片段化有所好转,以葛芪参妙组恢复效果最好。WB检测结果显示,各组药物干预后,Drp1蛋白有不同程度降低(P<0.05),其中以葛妙、葛芪参妙组中Drp1蛋白表达降低幅度最大(P<0.01),优于强的松组(P<0.01)。结论 葛根复方能够降低Drp1蛋白表达,减少线粒体分裂,促进线粒体恢复以及缓解骨骼肌组织的损伤。
Objective To investigate the effect of Kudzuvine root compound on Drp1 protein in skeletal muscle and mitochondria in experimental autoimmune myasthenia gravis(EAMG) rats. Methods The rats were randomly divided into 8 groups, with 10 rats in each group. Except for the blank group, the other groups were used for EAMG model construction using an artificial murine AchR-α subunit 97-116 peptide emulsifier. The corresponding drugs of the Radix Puerariae group were intragastrically administered every day, and the blank model group was intragastrically administered with normal saline once daily for 28 consecutive days. At the end of the treatment, the skeletal muscle tissues of rats were stained with hematoxylin-eosin(HE) method, the mitochondrial ultrastructure was observed by transmission electron microscopy, and the expression of Drp1 protein in mitochondria of rats was detected by Western blot.Results Compared with the model group, the skeletal muscle tissues of Kudzuvine root group and the prednisone group were disorganized, the nuclear deviation and inflammatory infiltration were reduced, the mitochondrial swelling and deformation, the cristae structure was unclear, and the fragmentation was improved, with the best recovery effect in the Kudzuvine Shenmiao group. The results of WB showed that Drp1 protein was decreased after drug intervention in each group(P<0.05), and the decrease of Drp1 protein expression was the most noteworthy in the Gemiao and Geqimiao groups(P<0.01), which was superior to that in the prednisone group(P<0.01).Conclusion Kudzuvine root compound can reduce the expression of Drp1 protein, reduce mitochondrial division, promote mitochondrial recovery and alleviate skeletal muscle injury.
