摘要
为从蛋白质水平解析卷烟接装纸赋甜技术对人体产生的影响,采用串联质谱标签(TMT)标记的定量蛋白质组学技术对收集的不抽吸对照组、抽吸赋甜卷烟组以及抽吸未赋甜卷烟组唾液中的蛋白进行鉴定和分析。结果表明:(1)从3组样本中共鉴定到2099个蛋白,其中,从对照组、赋甜卷烟组及未赋甜卷烟组均鉴定到2042个蛋白;(2)与对照组相比,抽吸未赋甜卷烟组共有107个差异表达蛋白,从抽吸赋甜卷烟组仅鉴定到5个差异表达蛋白。与赋甜卷烟组相比,从未赋甜卷烟组唾液中共鉴定到59个差异蛋白,其中,上调蛋白有55个,下调蛋白有4个;(3)GO富集分析和KEGG通路分析表明,与赋甜卷烟组相比,未赋甜卷烟组差异表达蛋白在细胞对化学刺激响应相关的生物过程以及与有机酸结合的分子功能等方面显著富集,主要涉及代谢途径和药物代谢其他酶通路等。上述结果表明抽吸未赋甜卷烟对口腔产生较大的刺激,而抽吸赋甜卷烟对口腔影响明显减少。
To clarify the effects of smoking cigarette with sweetened tipping paper on the smokers,saliva samples were collected from the control group(KB)and the groups smoking cigarettes with sweetened tipping paper(SK-A)and cigarettes with unsweetened tipping paper(SK-B).The protein levels in the saliva samples were identified and analyzed with quantitative proteomics technology labeled by tandem mass tag(TMT).The results showed that:1)A total of 2099 proteins were identified in the samples from the three groups,of which 2042 proteins were identified in the samples from KB,SK-A and SK-B separately.2)Compared with KB,there were 107 and 5 differentially expressed proteins identified in the samples from SK-B and SK-A,respectively.Compared with SK-A,there were 59 differentially expressed proteins in the saliva samples from SK-B,including 55up-regulated proteins and 4 down-regulated proteins.3)Further bioinformatics analysis was conducted to classify the proteins by gene ontology(GO)and KEGG database.The differentially expressed proteins in SK-B vs SK-A samples were significantly enriched in the biological process of cellular response to chemical stimulus and the molecular function of organic acids binding,which mainly involved the metabolic pathways and drug metabolism-other enzymes pathways.These results suggested that the oral stimulation responses of smokers to cigarettes with unsweetened tipping paper were greater than those to cigarettes with sweetened tipping paper.
