GSPT1在膀胱癌组织中的表达及对STAT3信号的影响研究

摘要 目的探讨GSPT1在膀胱癌组织中的表达及对STAT3信号的影响。方法测定2015至2021年69例膀胱癌组织及癌旁组织GSPT1水平,分析其与膀胱癌临床病理关系。同时设T24膀胱癌细胞组、GSPT1(低表达)inhibitor组、GSPT1(过表达)mimics组,培养72 h候后,测定细胞增殖、侵袭、凋亡水平,RT-PCR法及蛋白印迹法测定各组细胞GSPT1、STAT3、JAK2水平。结果膀胱癌组中GSPT1蛋白表达阳性率高于癌旁组(P<0.01)。GSPT1蛋白表达与年龄相关性不明显(P>0.05);与TMN分期、病理学分级、复发、转移相关性明显,且TNM分期越高、病理学分期越高、有复发、有转移,GSPT1蛋白阳性表达率越高(P<0.01)。与T24膀胱癌细胞组比较,GSPT1 inhibitor组OD值、存活率、克隆形成数目、迁移率、穿膜数、GSPT1、STAT3、JAK2mRNA蛋白水平降低(P<0.01),GSPT1 mimics组OD值、存活率、克隆形成数目、迁移率、穿膜数、GSPT1、STAT3、JAK2mRNA蛋白水平升高(P<0.01);与GSPT1 inhibitor组比较,GSPT1 mimics组OD值、存活率、克隆形成数目、迁移率、穿膜数、GSPT1、STAT3、JAK2mRNA蛋白水平升高(P<0.01)。与T24膀胱癌细胞组比较,GSPT1 inhibitor组细胞凋亡率升高(P<0.01),GSPT1 mimics组细胞凋亡率降低(P<0.01);与GSPT1 inhibitor组比较,GSPT1 mimics组单细胞凋亡率降低(P<0.01)。结论GSPT1在膀胱癌组织上调,与TNM分期、病理学分级、复发、转移相关;GSPT1激活STAT信号通路促进膀胱癌的转移和侵袭。 Objective To investigate the expression of GSPT1 in bladder cancer tissues and its effects on on STAT3 signal.Methods The levels of GSPT1 in 69 cases of bladder cancer tissues and adjacent tissues collected from in our hospital from 2015 to 2021 were detected,and the its correlation with clinical pathological indexes was analyzed.Moreover the bladder cancer cells(T24)were cultured in vitro,which were divided into T24 bladder cancer cell group,GSPT1(low expression)inhibitor group,and GSPT1(overexpression)mimics group.After 72-hour culture,the levels of cell proliferation,invasion and apoptosis were measured.RT-PCR and Western Blot were used to detect the levels of GSPT1,STAT3,JAK2 in the cells.Results The positive rate of GSPT1 protein expression in bladder cancer group were significantly higher than those in para-cancerous tissue group(P<0.01).GSPT1 protein expression was not correlated with patient’s age(P>0.05),however,which was significantly correlated with TMN staging,pathological grade,recurrence,and metastasis,and the higher the TMN stage,the higher the pathological stage and the recurrence as well as the metastasis,and the higher the positive rate of GSPT1 protein expression(P<0.01).Compared with those in T24 bladder cancer cell group,the OD value,survival rate,number of clone formation,migration rate,number of transmembrane,GSPT1,STAT3,and JAK2 mRNA protein levels in GSPT1 inhibitor group were significantly decreased(P<0.01),however,which in GSPT1 mimics group were significantly increased(P<0.01).Compared with that in T24 bladder cancer cell group,the apoptosis rate in GSPT1 inhibitor group was significantly increased(P<0.01),however,which in GSPT1 mimics group was significantly decreased(P<0.01).Compared with that in GSPT1 inhibitor group,the single cell apoptosis rate in GSPT1 mimics group was significantly decreased(P<0.01).Conclusion The expression levels of GSPT1 are up-regulated in bladder cancer tissues,which is related to TMN staging,pathological grade,recurrence and metastasis,and GSPT1 may promote the metastasis and invasion of bladder cancer by activating STAT3/Jak2 signaling pathway.
