摘要
目的:研究调和益胃颗粒对乙醇诱导小鼠胃溃疡保护作用及其相关机制。方法:采用乙醇诱导小鼠建立胃溃疡模型,采用HE染色观察各组小鼠胃组织病理学改变,采用生化试剂盒检测各组小鼠血清中丙二醛(MDA)、髓过氧化物酶(MPO)、超氧化物歧化酶(SOD),采用ELISA法测定各组小鼠血清细胞因子白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量,采用蛋白印迹法检测胃组织中p-NF-κBP65和p-IκBα蛋白表达。结果:调和益胃颗粒可有效减轻乙醇所致小鼠胃溃疡病理学变化,显著提高血清中SOD水平,并能降低MDA及MPO水平,并同时降低IL-6和TNF-α含量,减少胃组织p-NF-κBp65和p-IκBα的蛋白表达水平。结论:调和益胃颗粒对乙醇诱导的急性胃溃疡有保护作用,其机制与抗炎与抗氧化相关。
Objective:To determine the protective effect of Tiaoheyiweikeli on gastric ulcer induced by ethanol in mice.Methods:The BABL/C mice were randomly allocated into control,ethanol,omeprazole(20 mg/kg),Tiaoheyiweikeli(0.5 g/kg)and Tiaoheyiweikeli(1 g/kg).The blood samples were analyzed for the superoxide dismutase(SOD),methane dicarboxylic aldehyde(MDA),myeloperoxidase(MPO),interleukin-6(IL-6)and tumor necrosis factor alpha(TNF-α).Besides,the stomach samples were taken for histologic assessment.The protein levels of p-NF-κBP65,p-IκBαwere detected by western blot analysis.Results:Pretreatment with Tiaoheyiweikeli prior to the administration of ethanol significantly decreased the activity of MDA and the concentrations of IL-6 and TNF-αin serum.The expressions of p-NF-κBP65 and p-IκBαwere also reduced by Tiaoheyiweikeli with the expressions of NF-κBP65 and IκBαas inner controls.At the same time,pretreatment with Tiaoheyiweikeli also significantly improved the vitality of SOD and MPO in stomach.Conclusion:The findings revealed the anti-inflammatory effects of Tiaoheyiweikeli(0.5 g/kg,1 g/kg)against the ethanol-induced gastric ulcer.
