摘要
目的:探讨不同造模方法对大鼠蛛网膜下腔出血(SAH)后的脑部血流变化的影响。方法:分别用颈动脉血管穿孔(EVP)、单次自体动脉血注射(SI)和双次自体动脉血注射(DI)三种方法制备大鼠SAH模型,颅内压(ICP)检测仪检测大鼠ICP,激光多普勒血流仪检测大鼠局部血流量(rCBF),HE染色检测基底动脉痉挛,并比较不同造模方法对大鼠死亡率的影响。结果:EVP组大鼠ICP高于SI组和DI组,同时DI组ICP高于SI组。EVP组大鼠rCBF小于SI组和DI组。EVP组大鼠基底动脉痉挛严重程度大于DI组和SI组,同时,DI组大鼠基底动脉痉挛程度大于SI组。此外,EVP组大鼠死亡率高于DI组和SI组。结论:EVP模型大鼠脑部血流量严重减少,更适合于研究SAH早期脑血流变化。
Objective: To investigate the effect of different modeling methods on cerebral blood flow changes after subarachnoid hemorrhage(SAH) in rats. Methods: The rat SAH model was prepared by three methods: endovascular perforation(EVP), single injection of autologous blood into the cisterna magna(SI) and double injection of autologous blood into the cisterna magna(DI). Intracranial pressure(ICP) was measured continuously using an ICP sensor. The regional local blood flow(rCBF) was measured using laser Doppler flowmeter. The basilar artery spasm was detected by hematoxylin and eosin(HE) staining. The effects of different modeling methods on the mortality of rats were compared. Results: ICP of EVP group was higher than that of SI group and DI group;The ICP of DI group was higher than that of SI group. The rCBF of EVP group was lower than that of SI group and DI group. The severity of basilar artery vasospasm in EVP group was more serious than that in DI group and SI group;Meanwhile, basilar artery vasospasm in DI group was more serious than that in SI group. In addition, the mortality of EVP group was higher than that of DI group and SI group. Conclusion: Cerebral blood flow was significantly reduced in EVP model rats, and this model is more suitable to study the rCBF changes in the early stage of SAH.
