毛蕊花糖苷对中波紫外线所致小鼠皮肤损伤的保护作用研究 被引量:3

Study on the Protective Effect of Verbascoside on the Skin Damage Induced by UVB in Mice

摘要 目的 研究毛蕊花糖苷对中波紫外线(UVB)照射所致小鼠皮肤损伤的保护作用。方法 将Balb/c小鼠随机分为正常对照组、UVB模型组、基质+UVB组、1%毛蕊花糖苷+UVB组、5%毛蕊花糖苷+UVB组、5%维生素E+UVB组,每组10只。除正常对照组外,其余4组小鼠采用UVB照射建立皮肤损伤模型,单次照射强度为0.326 mW·cm^(-2),每次照射10 min,隔日照射1次,共照射10次,累积照射剂量为1 956 mJ·cm^(-2)。基质组和各给药组小鼠分别于每次照射前30 min将不含药物的空白基质、1%毛蕊花糖苷乳膏、5%毛蕊花糖苷乳膏、5%的维生素E乳膏涂于小鼠背部脱毛处。末次照射24 h后,处死小鼠,取背部皮肤进行相关指标的检测:皮肤称质量及皮肤厚度测量检测皮肤水肿;苏木精-伊红(HE)染色观察小鼠皮肤组织形态学特征;免疫组织化学法检测小鼠皮肤组织磷酸化的H2AX组蛋白(Phosphorylated histone H2AX,γH2AX)的表达;酶联免疫吸附法测定皮肤组织中环丁烷嘧啶二聚体(cyclobutyl pyrimidine dimers,CPDs)、6-4光产物(6-4photoproducts, 6-4PPs)和8-羟基-2-脱氧鸟嘌呤核苷(8-hydroxy-2-deoxyguanosine, 8-OHdG)的水平。结果 与正常对照组比较,UVB模型组小鼠皮肤出现水肿、粗糙、红肿、溃烂、表皮增厚等;γH2AX表达量明显升高(P<0.01);CPDs、6-4PPs、8-OHdG的含量均明显增加(P<0.01)。与UVB模型组比较,毛蕊花糖苷给药组小鼠皮肤组织的水肿、粗糙、红肿、溃烂、表皮增厚等症状明显改善;组织中γH2AX的表达明显降低(P<0.01);CPDs、6-4PPs、8-OHdG的含量明显减少(P<0.01)。结论 毛蕊花糖苷能改善UVB照射引起的小鼠皮肤损伤,且这种作用的发挥可能与其降低CPDs、6-4PPs和8-OHdG等因子的表达,改善DNA损伤有关。 Objective To investigate the protective effect of verbascoside on the skin damage induced by ultraviolet B(UVB)irradiation in mice.Methods Balb/c mice were randomly divided into control group, UVB model group,vehicle + UVB group,1% verbascoside + UVB group,5% verbascoside + UVB group,and 5% vitamin E + UVB group,with 10 mice in each group. Except for the control group,mice of other groups were irradiated with UVB to establish skin injury models,with an intensity of single irradiation of 0.326 mW·cm^(-2),10 minute of each irradiation.Mice were exposed to UVB every other day for ten times,with a cumulative irradiation dose of 1 956 mJ·cm^(-2). The bare skin on the back of mice in the vehicle group and each administration group were treated with a drug-free blank vehicle, 1% verbascoside cream, 5% verbascoside cream and 5% vitamin E cream at 30 minutes before each irradiation. 24 hour after the last irradiation,the mice were sacrificed,and skin sliced from the back of mice was used for detection of related indicators:skin weighing and skin thickness measurement to detect skin edema. The morphological characteristics of skin tissue were observed by HE staining. The expression of phosphorylated histone H2AX(γH2AX)was detected by immunohistochemistry. The levels of cyclobutyl pyrimidine dimers(CPDs),6-4photoproducts(6-4PPs)and 8-hydroxy-2-deoxyguanosine(8-OHdG)were determined by ELISA.Results Compared with control group,the skin of mice in the UVB model group significantly showed edema,roughness,redness,ulceration,and epidermic thickening. The expression level of γH2AX was significantly increased(P<0.01)and the contents of CPDs,6-4PPs and 8-OHdG were significantly enhanced(P<0.01). Compared with UVB model group,the symptoms of edema,roughness,redness,ulceration,and epidermic thickening of the skin tissue of mice in each administration group were significantly improved. The expression of γH2AX was decreased(P<0.01)and the contents of CPDs,6-4PPs and 8-OHdG were also reduced(P<0.01).Conclusion Verbascoside can relieve the skin damage of mice caused by UVB irradiation. This effect may be related to suppression of the expressions of CPDs,6-4PPs and 8-OHdG,as well as alleviating DNA damage.
