摘要
[目的]探究水和乙醇对迷宫栓孔菌(Trametes gibbosa)的提取效果及其提取物的抗氧化活性,并分析其主要抗氧化成分。[方法]以水和60%乙醇为溶剂提取迷宫栓孔菌子实体,以DPPH自由基清除率、超氧阴离子自由基清除率以及铁离子还原力进行抗氧化活性评价,福林酚法测定多酚含量,硫酸蒽酮法测定多糖含量,将抗氧化指标与多酚和多糖含量之间进行Pearson相关性分析。[结果]水提取物的得率为10.60%,60%乙醇提取物得率为4.27%;水提取物的多酚含量较高,60%乙醇提取物的多糖含量较高;所有处理均表现出良好的DPPH自由基清除能力和铁离子还原力,以及一定程度的超氧阴离子自由基清除能力,且水提取物的前2项指标均显著高于60%乙醇提取物;3项抗氧化指标均与多酚和多糖含量呈极显著正相关。[结论]迷宫栓孔菌的水提取物和60%乙醇提取物均具有抗氧化活性,水提取物的抗氧化活性更强,多酚及多糖都是其提取物的重要抗氧化活性成分,多酚的活性更强。
[Objective]To explore the extraction effect of water and ethanol on Trametes gibbosa and the antioxidant activity of their extracts,and analyze its main antioxidant components.[Method]Water and 60%ethanol were used as the solvent to extract the fruiting body of Trametes gibbosa.The antioxidant activity was evaluated by DPPH radical scavenging rate,superoxide anion radical scavenging rate and iron ion reducing power.The polyphenol content was determined by Folin phenol method,and the polysaccharide content was determined by anthrone sulfate method.Pearson correlation analysis was conducted between the antioxidant index and the polyphenol and polysaccharide content.[Result]The yield of water extract was 10.60%,and the yield of 60%ethanol extract was 4.27%.The polyphenol content of the water extract was higher,the polysaccharide content of the 60%ethanol extract was higher.All treatments showed good DPPH radical scavenging and reducing capacity,and a certain degree of superoxide anion radical scavenging capacity,and the first two indicators of water extract were significantly higher than that of 60%ethanol extract;the three antioxidant indexes were significantly correlated with the contents of polyphenols and polysaccharides.[Conclusion]Both water and 60%ethanol extracts of Trametes gibbosa have antioxidant activity,but the water extract has stronger antioxidant activity.Both polyphenol and polysaccharideis are important active component of extracts from Trametes gibbosa,but polyphenols are more active.
