摘要
[目的]利用脂多糖(LPS)诱导急性氧化应激损伤,评价蛹虫草对机体氧化和抗氧化系统平衡的保护作用。[方法]KM系小鼠分别腹腔注射6、8、10 mg/kg LPS水溶液0.5 mL/只(n=5),确定LPS诱导氧化应激的最佳剂量和时间效应。试验组以推荐剂量0.61 mg/g的蛹虫草水溶液,对照组为同等剂量蒸馏水,连续灌胃28 d后,腹腔注射10 mg/kg LPS的2、4、6、8 h取材(n=5),分别记录饮食、饮水及脏器指数变化。[结果]10 mg/kg LPS引起的小鼠扎堆、精神萎靡、嗜睡、竖毛及腹泻等应激症状最为稳定。对照组在LPS注射后肝脏、肾脏、脑组织均出现T-AOC指数和CAT活性的急剧下降,MDA和H_(2)O_(2)含量明显增加,试验组各指标变化趋势较为温和,并早于对照组恢复至空白组水平。[结论]短时间内连续服用蛹虫草可以降低LPS诱导的氧化应激损伤,作为天然抗氧化添加剂具有一定的开发潜力。
[Objective]To evaluate the protective effect of Cordyceps militaris on the balance of oxidative and antioxidant systems by lipopolysaccharide(LPS)induced acute oxidative stress injury.[Method]KM mice were intraperitoneally injected with 6,8 and 10 mg/kg LPS aqueous solution 0.5 mL/animal(n=5),which determined the optimal dose and time effect of LPS induced oxidative stress.Then the experimental group was treated with 0.61 mg/g Cordyceps militaris aqueous solution and the control group was treated with distilled water.After continuous gavage of 0.5 mL/animal for 28 days,samples were collected at 0,2,4,6 and 8 hours after intraperitoneal injection of 10 mg/kg LPS(n=5).The changes of diet,drinking water and organ index were recorded.[Result]10 mg/kg LPS caused the most stable stress symptoms in mice,such as pile up,depression,drowsiness,bristling and diarrhea.After LPS injection,T-AOC index and CAT activity of liver,kidney and brain tissues in the control group decreased sharply,and MDA and H_(2)O_(2) content increased significantly.The change trend of each index in the test group was relatively mild,and recovered to the level of the blank group earlier than that in the control group.[Conclusion]Continuous administration of Cordyceps militaris in a short period of time can reduce LPS induced oxidative stress injury,which has certain development potential as a natural antioxidant additive.
