摘要
目的 基于液相色谱-质谱联用(LC-MS)研究健脾化浊调脂方对动脉粥样硬化(AS)小鼠肝脏内源性代谢物的调节机制。方法 采用高胆碱饮食诱导小鼠AS模型,观察小鼠肝脏组织病理变化,运用LC-MS技术分析小鼠肝脏样品代谢轮廓,结合主成分分析(PCA)、正交最小偏二乘判别分析(OPLS-DA)等手段鉴定并筛选出小鼠肝脏差异代谢物及代谢通路。结果 健脾化浊调脂方可改善AS小鼠肝脏脂质蓄积情况,并回调12种与AS相关的潜在生物标志物以及6条代谢通路(甘油磷脂代谢、嘌呤代谢、β-丙氨酸代谢、泛酸和辅酶A生物合成、组氨酸代谢以及苯丙氨酸代谢)。结论 健脾化浊调脂方可能通过干预AS小鼠肝脏脂质代谢、氨基酸代谢以及能量代谢等过程发挥抗AS作用。
Objective Based on liquid chromatography-mass spectrometry(LC-MS),the regulatory mechanism of Jianpi Huazhuo Tiaozhi Recipe on endogenous metabolites in liver of mice with atherosclerosis(AS) was studied.Methods The AS model induced by high choline diet was used to observe the pathological changes of mouse liver tissue.The metabolic profile of mouse liver samples was analyzed by LC-MS technique,and the differential metabolites and metabolic pathways of mouse liver were identified and screened by means of principal component analysis(PCA) and orthogonal least squares discriminant analysis(OPLS-DA).Results Jianpi Huazhuo Tiaozhi Recipe can improve the liver lipid accumulation of AS mice,and recall 12 potential biomarkers related to AS and 6 metabolic pathways(Glycerophospholipid metabolism,purine metabolism,beta-alanine metabolism,Pantothenate and CoA biosynthesis,histidine metabolism and phenylalanine metabolism).Conclusion Jianpi Huazhuo Tiaozhi Recipe can play an anti-AS role by intervening in the processes of lipid metabolism,amino acid metabolism and energy metabolism in the liver of AS mice.
