比较荧光分光光度法和紫外-可见分光光度法检测刺葡萄叶片总黄酮含量 被引量:4

Comparison of Fluorescence Spectrophotometry and UV-Vis Spectrophotometry for Determination of Total Flavonoids in Vitis davidii Leaves

摘要 根据黄酮类化合物能与铝离子形成稳定荧光络合物的原理,采用改进单纯形法优化最佳测定条件,提出基于荧光光谱的刺葡萄叶片总黄酮含量的测定方法,并与测定黄酮类化合物含量常用的紫外-可见分光光度法进行了比较。研究结果显示,荧光分光光度法测定刺葡萄叶片总黄酮含量的最佳条件为:pH值为4.9的乙酸-乙酸钠缓冲溶液2.5 mL,76%的乙醇,正丁醇为5.7 mL,5.7%的硝酸铝溶液为1.5 mL。本方法标准曲线的决定系数为0.999 6,回收率为100.72%,精密度的相对标准偏差为1.00%,重现性的相对标准偏差为1.16%,检出限为0.54μg/L。 According to the principle that flavonoids can form stable fluorescent complexes with aluminum ions, an improved simplex method was used to optimize the optimal measurement conditions, and a method for determination of total flavonoids from leaves of Vitis davidii based on fluorescence spectrum was established.The proposed method was compared with the ultraviolet-visible spectrophotometry commonly used for determination of flavonoids content.The results showed that the optimal conditions for the determination of total flavonoids in the leaves of Vitis davidii by fluorescence spectrophotometry were as follows: pH=4.9 acetic acid-sodium acetate buffer solution 2.5 mL,76% ethanol, 5.7 mL n-butanol, and 1.5 mL 5.7% aluminum nitrate solution.The coefficient of determination of the standard curve is 0.999 6,the recovery rate is 100.72%,the relative standard deviation of precision is 1.00%,the relative standard deviation of reproducibility is 1.16%,and the detection limit is 0.54 μg/L.
