基于UPLC指纹图谱和含量测定法评价不同干燥温度对羌活药渣质量的影响

摘要 目的:建立UPLC指纹图谱和含量测定方法,评价不同干燥温度下羌活水提药渣的质量。方法:测定羌活药渣UPLC图谱以计算各共有峰的残存量,并结合聚类分析(CA)和主成分分析(PCA)优选最佳处理温度。建立含量测定方法以同时测得不同样品中羌活醇和异欧前胡素的含量。结果:CA和PCA得到相似的结论,将样品划分为三类。PCA综合评分表明60℃为样品的最适烘干条件,与含量测定结果一致。结论:本研究提供了稳定且具有专属性的指纹图谱和含量测定方法,并作为羌活药渣质量评价和后续开发的参考基础。 Objective: To establish the UPLC fingerprint chromatogram and quantification method for water-extracted residues form Notopterygii Rhizome, and evaluate its quality by different drying temperatures. Methods: UPLC characterization was performed on Notopterygium residues so that to calculate remaining quantities of the common peaks, on which cluster analysis(CA) and principal component analysis(PCA) were carried out to select the optimal processing temperature. Meanwhile, content determination methods were set up for quantity calculation of notopterol and isoimperatorin simultaneously among various samples.Results: Results of CA and PCA shared the similar conclusion, in which all samples were classified into three groups. Moreover, comprehensive scoring by PCA suggested that 60 ℃ would be the optimal drying condition for the samples, which was consistent with outcomes from content analysis. Conclusion: The research provided both the stable and specified fingerprint chromatogram and content analysis methods, which enabled the study reference for the quality evaluation and future exploitation of Notopterygium residues.
