摘要
目的 研究上海市金山区宾馆及公共浴室水环境中嗜肺军团菌污染情况,并逐步建立本区嗜肺军团菌蛋白指纹图谱数据库。方法 2021年11月-12月对辖区8家宾馆和4家公共浴室采集淋浴水及相关环境样本,利用基质辅助激光解析电离-飞行时间质谱法对嗜肺军团菌进行快速鉴定及聚类分析。结果 宾馆淋浴水、淋浴水相关环境、公共浴室水样中,嗜肺军团菌检出率分别为35.00%(7/20)、25.00%(5/20)、30.00%(6/20),差异无统计学意义(P>0.05),检出嗜肺军团菌以LP3型为主,宾馆和公共浴室水环境细菌谱差异较大,宾馆淋浴水样中以常见的单胞菌和肠杆菌为主,公共浴室水样中以不动杆菌为主。嗜肺军团菌蛋白图谱聚类分析发现分为2个明显的分支。结论 辖区内宾馆及公共浴室水环境中存在明显的嗜肺军团菌污染,并且血清型种类多样,存在同一场所多种血清型共存的情况,需要引起重视,建议对相关场所进行定期监测并指导消毒,预防呼吸道感染的发生。利用基质辅助激光解析电离-飞行时间质谱法检测嗜肺军团菌可以大大缩短检测周期,提高检测准确度,还能对其进行聚类分析,建立蛋白指纹图谱数据库,为溯源提供依据。
Objective This paper aims to study the pollution of Legionella pneumophila in the water environment of hotels and public bathrooms in Jinshan District, Shanghai, and gradually establish a protein database of Legionella pneumophila. Methods From November to December 2021, samples of shower water and related environment were collected from 8 hotels and 4 public bathrooms. Matrix assisted laser analytical ionization-time of flight mass spectrometry(MALDI-TOF-MS) was used to rapidly identify Legionella pneumophila and make cluster analysis. Results The detection rates of Legionella pneumophila in hotel shower water, shower water-related environment, and public bathroom water were 35.00%(7/20), 25.00%(5/20), 30.00%(6/20). There was no statistical significance on the difference(P>0.05). The majority of the detected Legionella pneumophila was LP3. The bacterial spectrum of the water environment in hotels and public bathrooms was quite different, with monas and Enterobacteria the dominant in hotel shower water and Acinetobacter the dominant in public bathroom water. The cluster analysis of the Legionella pneumophila found that it was divided into two obvious branches. Conclu sion There is obvious pollution of Legionella pneumophila in the water environment of hotels and public bathrooms in Jinshan District, and there are various types of serotypes, and the coexistence of multiple serotypes in the same place requires attention. It is recommended that relevant places be regularly monitored and disinfected. Using MALDI-TOF MS to detect Legionella pneumophila can shorten the detection time, improve detection accuracy, and do cluster analysis, also can establish a protein fingerprint database to provide a basis for traceability.
