伊犁州薰衣草根际促生真菌的分离鉴定及活性探究

摘要 目的:从伊犁州薰衣草根际土壤中筛选出具有抑菌活性及抗氧化活性的真菌。方法:采用倍比稀释法和平板划线法进行薰衣草根际真菌的分离及纯化;以形态学和分子生物学方法对分离出的真菌进行鉴定;用乙酸乙酯萃取发酵产物并进行滤纸片抑菌分析;使用DPPH法及ABTS法比较发酵产物乙酸乙酯层和水层的抗氧化活性。结果:从伊犁州薰衣草根际土壤中分离得到一株真菌YSZ-6,经鉴定为卷枝毛霉;该菌对大肠埃希氏菌、金黄色葡萄球菌及枯草芽孢杆菌具有显著的抑菌活性;发酵产物乙酸乙酯层和水层都具有清除DPPH自由基及ABTS自由基的能力,且具有抗氧化活性的物质主要分布在乙酸乙酯层。结论:从伊犁州薰衣草根际分离纯化出一株根际真菌,为卷枝毛霉,具有一定的抑菌活性及抗氧化活性,本研究对薰衣草资源的综合利用具有一定的参考价值。 Objective:Screening fungi with antibacterial and antioxidant activities from the rhizosphere soil of Ili lavender.Methods:The lavender rhizosphere fungi are isolated and purified by dilution method and the plate streak method.The isolated fungi are identified based on the morphological and molecular biology methods.The fermentation product is extracted with ethyl acetate and the antibacterial analysis of filter paper.Using DPPH and ABTS method to compare the antioxidant activity of ethyl acetate layer and water layer of fermentation products.Results:A rhizosphere fungi(YSZ-6)is isolated from the rhizosphere soil of Ili lavender,which is identified as Mucor circinelloides.YSZ-6 showes antibacterial activity against Escherichia coli,Staphylococcus aureus and Bacillus subtilis.Antioxidant activity experiments found that both the ethyl acetate layer and the water layer of the fermentation product has the ability to scavenge DPPH and ABTS free radicals and the substances with antioxidant ability are mainly in ethyl acetate layer.Conclusion:A rhizosphere fungi is isolated and purified from the rhizosphere of Ili lavender,which is Mucor circinelloides,and has antibacterial and antioxidant activities.This study has certain reference value in the comprehensive utilization.
