荞麦花叶总黄酮对心律失常大鼠心肌细胞凋亡及Wnt/β-catenin/PPARγ通路的影响 被引量:6

Effect of total flavonoids of buckwheat flower and leaf on myocardial cell apoptosis and Wnt/β-catenin/PPARγpathway in arrhythmic rats

摘要 探讨荞麦花叶总黄酮对心律失常大鼠心肌细胞凋亡及Wnt/β-连锁蛋白(β-catenin)/过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptorγ,PPARγ)通路的影响。将SD大鼠随机分为对照组、模型组、荞麦花叶总黄酮低剂量(20 mg·kg^(-1))组、荞麦花叶总黄酮中剂量(40 mg·kg^(-1))组、荞麦花叶总黄酮高剂量(80 mg·kg^(-1))组、盐酸普萘洛尔(2 mg·kg^(-1))组,每组12只大鼠,通过舌下静脉注射1 mL·kg^(-1)的0.002%乌头碱,将除对照组外的其余各组大鼠制备为心律失常模型,以药物分组干预处理后,检测各组大鼠心律失常情况、心肌细胞凋亡、心肌组织谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)、丙二醛(malondialdehyde,MDA)及血清白细胞介素-6(interleukin-6,IL-6)、前列腺素E2(prostaglandin E2,PGE2)水平、心肌组织凋亡及Wnt/β-catenin/PPARγ通路相关蛋白表达。相比对照组,模型组大鼠心律失常评分、室性早搏次数、室颤持续时间、心肌细胞凋亡率、心肌组织MDA水平、血清IL-6及PGE2水平、心肌组织Bax、Wnt1及β-catenin蛋白表达水平显著升高,心肌组织GSH-Px及CAT水平、心肌组织Bcl-2、PPARγ蛋白表达水平显著降低;相比模型组,药物干预组大鼠心律失常评分、室性早搏次数、室颤持续时间、心肌细胞凋亡率、心肌组织MDA水平、血清IL-6及PGE2水平、心肌组织Bax、Wnt1及β-catenin蛋白表达水平降低,心肌组织GSH-Px及CAT水平、心肌组织Bcl-2、PPARγ蛋白表达水平升高,且荞麦花叶总黄酮各组呈剂量依赖性;荞麦花叶总黄酮高剂量组与盐酸普萘洛尔组相比,大鼠各指标水平差异无统计学意义。荞麦花叶总黄酮可抑制Wnt/β-catenin通路激活,上调PPARγ表达,减轻心律失常大鼠心肌组织氧化应激及炎症损伤,减弱心肌细胞凋亡,改善大鼠心律失常症状。 This paper aimed to investigate the effect of total flavonoids of buckwheat flower and leaf on myocardial cell apoptosis and Wnt/β-catenin/peroxisome proliferator-activated receptorγ(PPARγ)pathway in arrhythmic rats.SD rats were randomly divided into a control group,a model group,a low-dose(20 mg·kg^(-1))group of total flavonoids of buckwheat flower and leaf,a medium-dose(40 mg·kg^(-1))group of total flavonoids of buckwheat flower and leaf,a high-dose(80 mg·kg^(-1))group of total flavonoids of buckwheat flower and leaf,a propranolol hydrochloride(2 mg·kg^(-1))group,with 12 rats in each group.Except the control group,rats in other groups were prepared as models of arrhythmia by sublingual injection of 1 mL·kg^(-1)of 0.002%aconitine.After grouping and intervention with drugs,the arrhythmia,myocardial cells apoptosis,myocardial tissue glutathione peroxidase(GSH-Px),catalase(CAT),malondialdehyde(MDA),serum interleukin-6(IL-6),prostaglandin E2(PGE2)levels,myocardial tissue apoptosis,and Wnt/β-catenin/PPARγpathway-related protein expression of rats in each group were measured.As compared with the control group,the arrhythmia score,the number of ventricular premature beats,ventricular fibrillation duration,myocardial cell apoptosis rate,MDA levels in myocardial tissues,serum IL-6 and PGE2 levels,Bax in myocardial tissues,and Wnt1 andβ-catenin protein expression levels increased significantly in the model group,whereas the GSH-Px and CAT levels,and Bcl-2 and PPARγprotein expression levels in myocardial tissues reduced significantly.As compared with the model group,the arrhythmia score,the number of ventricular premature beats,ventricular fibrillation duration,myocardial cell apoptosis rate,MDA leve in myocardial tissues,serum IL-6 and PGE2 levels,Bax in myocardial tissues,and Wnt1 andβ-catenin protein expression levels reduced in the drug intervention groups,whereas the GSH-Px and CAT levels and Bcl-2 and PPARγprotein expression levels in myocardial tissues increased.The groups of total flavonoids of buckwheat flower and leaf were in a dose-dependent manner.There was no significant difference in the levels of each index in rats between the propranolol hydrochloride group and the high-dose group of total flavonoids of buckwheat flower and leaf.The total flavonoids of buckwheat flower and leaf inhibit the activation of Wnt/β-catenin pathway,up-regulate the expression of PPARγ,reduce oxidative stress and inflammatory damage in myocardial tissues of arrhythmic rats,reduce myocardial cell apoptosis,and improve the symptoms of arrhythmia in rats.
