基于网络药理学对前胡“降气化痰”功效的解释 被引量:4

Explaination of the Jiangqihuatan Efficacy of Peucedanum praeruptorum Dunn based on Network Pharmacology

摘要 目的:基于网络药理学研究方法初步阐述前胡“降气化痰”功效的科学内涵。方法:通过检索中药系统药理学数据库(TCMSP)对前胡潜在的活性成分和靶点进行初步预测;检索相关疾病数据库获得“咳、痰、喘”等疾病的靶点;将上述二者通过STRING网站获得的蛋白互作网络图进行分析;利用Metascape在线平台进行基因本体(GO)分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:前胡与“咳、痰、喘”等相关疾病的相交靶点63个,富集分析得到了包含血液循环、对外源性化合物刺激的反应、对激素反应、对无机物的反应、细胞对氮化合物的反应等生物学进程20个,其中涉及了TP53、JUN、AKT1、MAPK14、FOS等靶点;KEGG通路富集筛选得到了包含cAMP信号通路、VEGF信号通路、Fox O信号通路以及癌症等18条关键信号通路。结论:前胡通过多靶点、多通路发挥“降气化痰”功效,从而为前胡的新药研发以及临床合理用药提供了参考,也为相关疾病的治疗提供生物信息学基础。 Objective:To explain the scientific connotation of Jiangqihuatan efficacy of Qianhu by using Network Pharmacology research methods.Methods:Potential active compounds and targets were predicted by searching the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Relevant targets of“cough,phlegm,asthma”were predicted by searching relevant databases.The common targets of drug and diseases were analyzed by establishing protein-protein interaction network on the STRING website.Gene ontology(GO)analysis and Kyoto Genomics and Genomics Encyclopedia(KEGG)pathway enrichment analysis were carried out by the Metascape website.Results:Sixty-three common targets between Qianhu and the related diseases(cough,phlegm,asthma)were found.The obtained 20 biological processes including blood circulation,response to xenobiotic stimulus,response to hormone,response to inorganic substance,cellular response to nitrogen compound and so on,,which involved TP53,JUN,AKT1,MAPK14,FOS and other targets.The 18 key pathways,including cAMP signal pathway,VEGF signal pathway,FoxO signal pathway and pathway in cancer and so on,were screened via KEGG pathway enrichment analysis.Conclusion:The play of Jiangqihuatan efficacy of Qianhu may via multi-targets and multi-pathway.The research results provide references for new drug research and development,rational drug use in clinical.The results also provide bioinformatics basis for the treatment of relevant databases.
