摘要
目的通过计算机虚拟筛选技术和实验验证手段,探究气滞胃痛颗粒“达乙木,疏己土”的理论内涵。方法通过网络药理学筛选气滞胃痛颗粒(QZWT)的活性化合物,通过中药系统药理学数据库和分析平台(TCMSP)等数据库收集气滞胃痛颗粒中的活性成分靶点,检索DisGeNET数据库等疾病数据库,得到疾病靶点,取交集靶点,通过京都基因与基因组百科全书(KEGG)富集分析,探讨QZWT活性成分的潜在机制,构建中药-成分-靶点-通路网络图。通过建立碘乙酰胺(IA)诱导的功能性消化不良大鼠模型,空白组、模型组予等体积无菌蒸馏水灌胃,多潘立酮组、QZWT组分别予对应浓度的药物灌胃10 d。苏木素-伊红(HE)染色观测十二指肠病理变化,酶联免疫吸附测定法(ELISA)检测血清白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,蛋白免疫印迹(Western blot)、免疫荧光检测表皮生长因子受体(EGFR)、闭锁蛋白1(ZO-1)、闭合蛋白(Occludin)表达水平。结果与空白组比较,模型组血浆炎症因子IL-6、IL-1β、TNF-α水平升高(P<0.05),十二指肠组织ZO-1、Occludin表达下调,EGFR表达上调(P<0.05);与模型组比较,QZWT组血浆炎症因子IL-6、IL-1β、TNF-α水平降低(P<0.05),十二指肠组织ZO-1、Occludin表达上调,EGFR表达下调(P<0.05)。结论以“达乙木,疏己土”的理论为指导,气滞胃痛颗粒通过抗炎、修复肠道屏障治疗功能性消化不良。
Objective To explore the theoretical connotation of Qizhi Weitong Granule(QZWT,气滞胃痛颗粒)“regulating Yimu,dredging Jitu”by means of computer virtual screening and experimental verification.Methods The active compounds of QZWT were screened through network pharmacology,and the targets of the active ingredients in QZWT were collected through pharmacological database and analysis Platform of TCM System(TCMSP),and the disease targets were obtained by searching DisGeNET database and other disease databases,and the intersection targets were obtained.The potential mechanism of QZWT active components was explored by enrichment analysis of Kyoto Gene Genome Encyclopedia(KEGG),and the network diagram of QZWT-component-target-pathway was constructed.FD model induced by iodoacetamide was established.Blank group and model group were intragastric with equal volume of sterile distilled water,domperidone group and QZWT group were intragastric with corresponding concentration for 10 days.Pathological changes of duodenum were observed by hematoxylin-eosin(HE)staining,and serum levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were determined by enzyme-linked immunosorbent assay(ELISA).The expression levels of epidermal growth factor receptor(EGFR),Zona occludens protein 1(ZO-1)and Occludin were detected by Western blot and immunofluorescence.Results Compared with blank group,plasma levels of inflammatory factors IL-6,IL-1βand TNF-αwere increased in model group(P<0.05).The expression of ZO-1 and Occludin in duodenum was down-regulated,and the expression of EGFR was up-regulated(P<0.05).Compared with model group,the levels of plasma inflammatory factors IL-6,IL-1βand TNF-αin QZWT group were decreased(P<0.05),the expression of ZO-1 and Occludin in duodenum was up-regulated,while EGFR was down-regulated(P<0.05).Conclusion Guided by the theory of“regulating Yimu,dredging Jitu”,Qizhi Weitong Granule treating functional dyspepsia through anti-inflammatory and repairing intestinal barrier.
