叶面喷施脱落酸对黄瓜果实品质的影响 被引量:1

摘要 【目的】探讨叶面喷施脱落酸(ABA)对黄瓜品质的影响,为外源施用激素ABA调控黄瓜果实品质提供生产指导和理论基础。【方法】以‘川翠13号’黄瓜品种为试验材料,通过叶面喷施不同浓度的ABA(0、50、100、200和400μmol/L),研究其对黄瓜果实的可溶性糖、抗坏血酸以及风味物质反,顺-2,6-壬二烯醛累积的影响。【结果】叶面喷施50~200μmol/L ABA提高了黄瓜果实中可溶性糖和抗坏血酸含量,降低了风味物质的累积。当叶面喷施400μmol/L的ABA时,抗坏血酸含量显著降低。为了进一步分析外源施用ABA影响黄瓜果实品质的机理,对黄瓜果实进行转录组分析,ABA处理叶片后,果实差异表达基因中上调的有94个,下调的有75个,GO基因功能富集分析结果表明,大量差异基因参与转录调控以及大分子的生物合成调控过程,KEGG通路富集分析结果表明差异表达基因主要参与代谢途径和激素信号转导途径,说明外源施用ABA可能通过调控黄瓜果实代谢过程以及激素信号转导过程影响果实的品质。此外,44个转录因子的表达也受到外源ABA的诱导,主要包括bHLH、bZIP、FAR1、MYB以及NAC等转录因子。【结论】本研究通过对黄瓜果实品质以及转录水平变化进行分析,发现叶片喷施ABA通过转录水平调控相关基因的表达正向调控可溶性糖、抗坏血酸含量,负向调控黄瓜风味物质的累积。 【Objective】The present paper investigated the effects of ABA on the fruit quality of cucumber to offer operation instruction and theoretical basis in the fruit improvement via exogenous ABA.【Method】Huabei cucumber variety‘Chuancui No.13’was used as materials and the method of foliar spraying was used.The effects of spraying different concentration of ABA(0,50,100,200 and 400μmol/L)on the soluble sugar,ascorbic acid and flavor substance trans⁃2,cis⁃6⁃Nonadienal of the cucumber fruit were studied.【Result】Foliar spraying 50-200μmol/L ABA promoted the accumulation of soluble sugar and ascorbic acid content,but decreased the content of flavor sub⁃stance.Foliar spraying 400μmol/L ABA decreased the ascorbic acid content significantly.To further analyze mechanism of ABA influencing the quality of cucumber,RNA⁃seq was performed to analyze the cucumber fruit.A total of 169 differentially expression genes(DEGs,94 genes were up⁃regulated,and 75 genes were down⁃regulated)were found after the foliar ABA application in the cucumber fruit compared to the control.Gene ontology(GO)enrichment showed that multiple DEGs were involved in transcription regulation and regulation of macromol⁃ecule biosynthetic process.KEGG pathway enrichment showed that most DEGs participated in biosynthesis and hormone signaling pathway,suggesting that exogenous ABA affected cucumber fruit quality via macromolecule biosynthesis and hormone signaling pathway.In addition,expression level of 44 transcription factors was influenced by exogenous ABA application,including TFs from families of bHLH,bZIP,FAR1,MYB and NAC.【Conclusion】The research investigated effect of foliar spraying ABA on quality and transcriptome profiling changes of cucumber fruit and proposed that exogenous ABA affected genes expression at transcription levels to positively regulate soluble sugar and as⁃corbic acid content and negatively regulate accumulation of flavor substance.
