摘要
目的:观察冰黄五苓散直肠滴注对于大脑中动脉闭塞(MCAO)模型大鼠的血清神经元特异性烯醇化酶(NSE)、S100β蛋白、脑组织水孔蛋白4(AQP4)的影响。方法:将90只SD大鼠随机分为假手术组、模型组、冰黄五苓散组。对各组大鼠不同时间点的神经功能缺损进行评分,采用干-湿重法测定脑组织含水量,采用酶联免疫吸附试验(ELISA)检测血清NSE、S100β蛋白水平,采用免疫组织化学法检测脑组织AQP4表达。结果:冰黄五苓散组神经功能缺损评分、脑组织含水量明显低于模型组(P<0.05);冰黄五苓散组血清NSE、S100β蛋白、脑组织AQP4明显低于模型组(P<0.05)。结论:冰黄五苓散能够改善MCAO模型大鼠脑水肿情况,起到保护神经功能的作用。
Objective:To observe the effect of rectoclysis with Binghuang Wuling Powder on neuron-specific enolase(NSE),S100β,and aquaporin 4(AQP4)in rats with middle cerebral artery occlusion(MCAO).Methods:Ninety SD rats were randomly assigned into a sham operation group,a model group,and a Binghuang Wuling Powder group.The rats in each group were scored for neurological deficits at different time points.The brain water content was measured by dry-wet weight method.The serum levels of NSE and S100βwere determined by enzyme-linked immunosorbent assay(ELISA),and AQP4 expression in the brain tissue by immunohistochemistry.Results:The Binghuang Wuling Powder group had lower neurological deficit score,brain water content,serum NSE and S100βlevels,and AQP4 expression level than the model group(P<0.05).Conclusion:Binghuang Wuling Powder can mitigate cerebral edema and protect nerve function in the treatment of MCAO rats.
