摘要
目的:探讨迷迭香酸(RA)对肺癌95D和A549细胞的凋亡作用及其作用机制。方法:CCK-8检测RA(25、50、100、200、300、400μmol/L)对95D和A549细胞不同时间(42、72 h)的细胞增殖作用;碘化丙啶(PI)单染法检测RA(150、200、250μmol/L)对95D和A549细胞周期的影响;异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)凋亡试剂盒检测RA(150、200、250μmol/L)对95D和A549细胞凋亡的影响;Western Blot检测RA(150、200、250μmol/L)对95D和A549细胞中p53和p21蛋白水平的影响。结果:95D和A549细胞随着RA给药剂量增加细胞存活率逐渐降低;RA各剂量组95D和A549细胞较空白对照组细胞阻滞在G1/G0期(P<0.05,P<0.01);RA高剂量组95D早凋亡细胞和A549中早、晚凋亡细胞凋亡量较空白对照组显著升高(P<0.01);与空白对照组比较,RA各剂量组95D和A549细胞中p53和p21蛋白表达水平升高。结论:RA可以抑制95D和A549细胞的增殖,周期阻滞G1/G0期,进而诱导细胞凋亡,其机制可能与调节p53/p21信号通路有关。
Objective:To study the effect of rosmarinic acid(RA)on lung cancer 95D and A549 cells and its mechanism.Methods:Cell proliferation and activity assay-8(CCK-8)was used to detect the proliferation of RA(25,50,100,200,300,400μmol/L)on 95D and A549 cells at different time(42,72 h).The effects of RA(150,200,250μmol/L)on 95D and A549 cell cycles were detected by PI staining.Annexin V-FITC/PI apoptosis kit was used to detect the effects of RA(150,200,250μmol/L)on apoptosis of 95D and A549 cells.Western Blot was used to detect the effects of RA(150,200,250μmol/L)on p53 and p21 protein levels in 95D and A549 cells.Results:The viability of 95D and A549 cells decreased gradually with the increase of RA dose.Compared with blank control group cells,95D and A549 cells in each RA dose groups were blocked in G,/Go phase(P<0.05,P<0.01).Compared with the blank control group,the amount of apoptosis of 95D early apoptotic cells and A549 medium early and late apoptotic cells was significantly higher in the RA high dose group(P<0.01).Compared with the blank control group,the protein level of p53 and p21 in each RA dose groups increased.Conclusion:RA can inhibit the proliferation of 95D and A549 cells,the cell cycle is arrested in G,/Go phase,and induces apoptosis,which may be related to the regulation of p53/p21 signaling pathway.
