摘要
目的:考察比较八号烧伤膏和烧伤凝胶治疗大鼠深Ⅱ度烫伤效果。方法:大鼠随机分组,90℃铁棒烫伤建立大鼠深Ⅱ度烫伤模型。早晚两次创面涂药,首次给药后连续十个时间点眼眶取血,HPLC检测血液中没食子酸浓度;ELISA分析第14 d血浆中IL-6和IL-10水平;HE切片分析创面恢复情况。结果:造模后第19 d模型组痂全脱落,八号烧伤膏组第15 d痂全部脱落,而烧伤凝胶组大鼠创面痂未有脱落;切片分析表明八号烧伤膏组大鼠创面新生皮层结构紧密,优于烧伤凝胶组;八号烧伤膏和烧伤凝胶首次给药后8 h时没食子酸浓度均达到最高,且组间差异极显著(P<0.001),12 h时没食子酸浓度降至最高浓度一半;八号烧伤膏和烧伤凝胶均显著降低血浆中IL-6水平(P<0.05)和提高IL-10水平(P<0.01及P<0.05),且八号烧伤膏组IL-10水平显著高于烧伤凝胶组(P<0.05)。结论:八号烧伤膏透皮吸收及抑制炎症效果均好于烧伤凝胶,治疗烫伤效果优于烧伤凝胶。
Objective:To investigate and compare the effect of Bahao burn slurry and burn gel on the healing of scald wounds in rats.Methods:Male Sprague-Dawley rats were divided randomly into groups.Except control group,all other rats were established the deep second degree scald rat models by 90℃hot iron.Mice were respectively given pure water,gel,bahao burn slurry amd burn gel twice per day.HPLC analyzed the concentration of gallate in plasma of rat collected at consecutive points in time after the first time of drug therapy.The wound healing was recorded and analyzed through HE straining.IL-6 and IL-10 in plasma was detected by ELISA kits.Results:Compared with the fell of scabs in model group until the 19th day after model established,scabs in bahao burn slurry group rats fell off completely at the 15th day.And there was no scabs falling off in burn gel group during the whole process.HE straining analysis showed the new skin epithelium of bahao burn slurry group rats was more thick than gel group rats.HPLC analysis shown that the concentration of gallate in plasma reached the highest concentration after 8 hours,and the difference between the two groups was extremely significant(P<0.001).At 12 hours,the concentration of gallic acid dropped to half of the maximum concentration.Bahao burn slurry and burn gel treatments suppressed the elevation of IL-6(P<0.05)and increased the level of IL-10 significantly(P<0.01 and P<0.05)at the same time.And bahao burn slurry group had higher level of IL-10 than burn gel group(P<0.05).Conclusion:Compared with burn gel,bahao burn slurry has better effect on transdermal absorption and anti-inflammatory.These results indicated that bahao burn slurry has better effect to therapy scald than burn gel.
