摘要
通过网络药理学技术结合层次分析法(AHP)-熵权法及多指标正交试验优选川芎-天麻药对提取工艺。利用网络药理学及分子对接技术对川芎、天麻潜在活性成分及靶点进行筛选及验证,并参考2020年版《中国药典》,确定川芎-天麻药对的关键成分为天麻素、巴利森苷B、巴利森苷C、巴利森苷E、阿魏酸、丁苯酞。以各指标成分的提取量、出膏率作为综合评价指标,运用AHP-熵权法主客观组合赋权及正交试验设计,确定川芎-天麻药对最佳提取工艺参数:乙醇体积分数50%,料液比为1∶8(g·mL^(-1)),提取3次,每次1.5 h。通过网络药理学及分子对接技术确定工艺评价指标成分,优选的川芎-天麻药对提取工艺经验证,稳定且重复性好,可为后续的深入研究提供参考。
To optimize the extraction process of Chuanxiong Rhizoma-Gastrodia elata drug pair by network pharmacology combined with analytic hierarchy process(AHP)-entropy weight method and Multi-index Orthogonal Test.The potential active ingredients and targets of Chuanxiong Rhizoma-Gastrodia elata drug pair were screened by network pharmacology and molecular docking techniques,and the process evaluation indexes were determined with reference to the 2020 Edition of the Chinese Pharmacopoeia.The core components of the Chuanxiong Rhizoma-Gastrodia elata drug pair were determined as gastrodin,parishin B,parishin C,parishin E,ferulic acid and 3-butylphthalide.The optimal extraction process conditions were obtained by using the subjective and objective combination empowerment of AHP-entropy weight method and orthogonal test:ethanol volume fraction of 50%,solid-liquid ratio of 1:8(g·mL^(-1)),extraction for three times and two hours each time.Through network pharmacology and molecular docking technology,the process evaluation index was determined,and the preferred Chuanxiong Rhizoma-Gastrodia elata drug pair was reasonable and stable for the extraction process,which could provide reference for subsequent in-depth research.
