十二烷基苯磺酸钠暴露对黄颡鱼稚鱼的遗传毒性

Genotoxicity of Sodium Dodecyl Benzene Sulfonate Exposure to Juvenile Pelteobagrus fulvidraco

摘要 【目的】评估十二烷基苯磺酸钠(SDBS)暴露对黄颡鱼(Pelteobagrus fulvidraco)稚鱼的遗传毒性。【方法】将黄颡鱼稚鱼(孵化120 h)暴露于质量浓度分别为0、0.2、0.4、0.6、0.8 mg/L的SDBS中96 h后,采用微核实验和单细胞凝胶电泳技术测定不同暴露浓度的黄颡鱼稚鱼红细胞微核率和细胞尾部DNA损伤率,检测稚鱼组织中DNA氧化损伤的特异性产物8-羟基脱氧鸟苷的含量。【结果】黄颡鱼稚鱼红细胞微核率变化范围为(0.22±0.02)%~(0.59±0.03)%,且随着SDBS暴露浓度的升高,对应黄颡鱼稚鱼红细胞微核率也显著增加(P<0.05)。SDBS暴露对黄颡鱼稚鱼细胞均具有不同程度的遗传损伤,DNA损伤率在9.4%~24.7%。SDBS暴露浓度(x)与稚鱼细胞尾部DNA含量(y)呈现一定的剂量效应关系,其对应的回归方程为y^(^)=4.6x+1.48(R2=0.9396,n=24,P<0.05)。黄颡鱼稚鱼组织中8-羟基脱氧鸟苷质量浓度变化范围为(1.7±0.36)~(8.0±0.58)μg/L,随着SDBS暴露浓度的升高,对应黄颡鱼稚鱼组织中8-羟基脱氧鸟苷质量浓度也显著增加(P<0.05)。【结论】0.2~0.8 mg/L的SDBS暴露对黄颡鱼稚鱼具有遗传毒性。 【Objective】To assess the genotoxicity of sodium dodecyl benzene sulfonate(SDBS)exposure to juvenile Pelteobagrus fulvidraco.【Method】Juvenile P.fulvidraco(120 h of incubation)were exposed to SDBS at 0,0.2,0.4,0.6 and 0.8 mg/L mass concentration for 96 h,then micronucleus assay,and single cell gel electrophoresis(SCGE)were used to determine the erythrocyte and fish tail DNA damage rates of juvenile P.fulvidraco exposed to SDBS at different concentrations.The rate of micronuclei and the rate of DNA damage in the tail of fish cells were determined by SCGE and micronucleus assay.The mass concentration of 8-hydroxydeoxyguanosine,which is the specific product of oxidative DNA damage in the tissue of juvenile fish,was determined.【Results】The erythrocyte micronucleus rate of juvenile P.fulvidraco ranged from(0.22±0.02)%to(0.59±0.03)%,and increased significantly(P<0.05)with increasing SDBS exposure.There was a dose-effect relationship between SDBS exposure and tail DNA percentage of fish cells of juvenile fish,following the corresponding regression equation y^(^)=4.6x+1.48(R2=0.9396,n=24,P<0.05).Tissue 8-hydroxydeoxyguanosine mass concentration in juvenile P.fulvidraco ranged from(1.7±0.36)μg/L to(8.0±0.58)μg/L.As the concentration of SDBS exposure increased,the corresponding tissue 8-hydroxydeoxyguanosine mass concentration in juvenile P.fulvidraco also increased significantly(P<0.05).【Conclusion】0.2-0.8 mg/L SDBS exposure was genotoxic to juvenile P.fulvidraco.
