摘要
目的探讨乌梅丸对乳腺癌MDA-MB-231-GFP细胞小鼠移植瘤的作用及其可能机制。方法选择SPF级雌性小鼠为试验动物,建立人乳腺癌MDA-MB-231-GFP细胞小鼠移植模型,随机分为模型组(灌胃口服0.9%氯化钠溶液)、阳性对照组(腹腔注射紫杉醇20 mg/kg)、乌梅丸组(灌胃乌梅丸颗粒剂药液),每日1次,连续30 d。观察小鼠一般情况,利用荧光成像系统采集肿瘤图像,以系统自带软件计算肿瘤面积;称取小鼠肿瘤质量和肺癌转移灶数;qPCR检测小鼠肿瘤组织中Fas、FasL mRNA水平;Western blotting检测小鼠肿瘤组织MDAMB-231细胞中Fas相关死亡域蛋白(FADD)、活性半胱天冬酶8(cleaved caspase-8)、Fas、FasL蛋白表达。结果模型组、乌梅丸组和阳性对照组小鼠肿瘤面积持续增大,呈时间依赖性(P<0.05);与模型组比较,乌梅丸组与阳性对照组小鼠肿瘤面积较小,肿瘤质量较低,肺癌转移灶数量较少(P<0.05);与乌梅丸组比较,阳性对照组小鼠瘤面积较小,肿瘤质量较低,肺癌转移灶数量较少(P<0.05)。与模型组比较,乌梅丸组小鼠Fas mRNA相对表达量较高、FasL mRNA相对表达量较低,FADD、cleaved caspase-8、Fas蛋白相对表达量较高、FasL蛋白相对表达量较低(P<0.001)。结论乌梅丸对乳腺癌MDA-MB-231-GFP细胞小鼠移植瘤有抑制作用,并抑制肺转移,其可能机制与调控Fas/FasL通路蛋白有关。
Objective To investigate the eff ect of Wumei Pills on the transplanted tumor of breast cancer MDAMB-231-GFP cells in mice and its possible mechanism.Methods SPF grade female mice were selected as the experimental animals to establish a mouse transplantation model of human breast cancer MDA-MB-231-GFP cells.They were randomly divided into a model group(oral 0.9%sodium chloride solution by gavage),a positive control group(intraperitoneal injection of paclitaxel 20 mg•kg^(-1)),and a Wumei Pills group(Wumei Pills granule liquid by gavage),once a day for 30 consecutive days.The general conditions of the mice were observed,and their tumor images were collected with the fl uorescence imaging system,and the tumor areas were calculated with the system's built-in software.The mouse tumor masses and the number of lung cancer metastases were weighed.The mRNA levels of Fas and FasL in the mouse tumor tissues were detected with qPCR,and the protein expressions of the Fasrelated death domain protein(FADD),cleaved caspase-8,Fas and FasL in the MDA-MB-231 cells of mouse tumor tissues were detected with Western blotting.Results The tumor areas of the mice in the model group,the Wumei Pills group and the positive control group continued to increase in a time-dependent manner(P<0.05).Compared with the mice in the model group,the mice in the Wumei Pills group and the positive control group had a smaller tumor area,a lower tumor mass and fewer lung cancer metastases(P<0.05).Compared with the mice in the Wumei Pills group,the mice in the positive control group had a smaller tumor area,a lower tumor mass,and fewer lung cancer metastases(P<0.05).Compared with the model group,the relative mRNA expression of Fas in the Wumei Pills group was higher,while the relative mRNA expression of FasL was lower,the relative protein expressions of FADD,cleaved caspase-8 and Fas were higher,while the relative protein expression of FasL was lower(P<0.001).Conclusion Wumei Pills has an inhibitory eff ect on the breast cancer MDA-MB-231-GFP cell transplanted tumor in mice and can inhibit lung metastasis,whose possible mechanism is related to the regulation of Fas/FasL pathway proteins.
