摘要
目的分析长春市2016—2020年度甲型H1N1流感病毒血凝素(HA)基因特征,了解其变异情况及遗传进化特征。方法选取2016—2020年度甲型H1N1流感病毒分离株109株,PCR扩增HA基因并进行序列测定;利用生物信息学软件对分离株的HA基因进行进化和变异分析。结果长春市109株分离株与疫苗株A/California/07/2009的HA蛋白相比均有S84N、S162N和I216T共同的氨基酸位点变异,且有1株发生了HA蛋白D222N位点突变,提示此毒株可引起患者下呼吸道症状。109株病毒株的HA基因之间核苷酸同源性为96.9%~100%,氨基酸同源性为97.2%~100%。在进化树分析上,都属于6B.1大分支,并且随着时间的推移,HA在基因进化树上离早期疫苗株A/California/7/2009越来越远,同一年份里也有毒株位于不同的小分支内。结论长春市2016—2020年度甲型H1N1流感病毒血凝素(HA)基因在不断的变异中,应持续进行监测,为预防甲型H1N1流感大流行提供参考。
Objective To analyze the genetic characteristics of hemagglutinin(HA)gene of influenza A(H1N1)pdm09virus in Changchun during 2016-2020and to comprehend the variation and evolutionary characteristics.Methods Influenza A(H1N1)pdm09virus strains were selected from 2016-2020to perform sequencing of HA gene.And the variation and evolutionary characteristics of hemagglutinin(HA)gene were obtained with the bioinformatics software.Results The influenza A(H1N1)pdm09virus strains had some common mutation sites compared with vaccine strain A/California/07/2009.And there was one virus strain with D222Nmutation site.It suggested that the strain can cause lower respiratory symptoms.Sequence analysis showed that HA gene was highly homologous to vaccine strains.?C`?C?The phylogenetic analysis showed that HA gene clustered nearly on the identical phylogenetic tree,belonging to branch 6B.1.Conclusion The HA gene of influenza A(H1N1)pdm09virus strains circulated in Changchun from 2016to 2020 presented continual genetic variation.Continuous surveillance should be strengthened,in order to prevent pandemic of influenza A(H1N1)pdm09.
