摘要
目的探讨亚甲基四氢叶酸脱氢酶2(MTHFD2)在非肌层浸润性膀胱癌(NMIBC)中的表达及意义。方法选取NMIBC患者82例,均采用免疫组化染色法检测MTHFD2的表达状况。分析MTHFD mRNA于NMIBC组织内的表达状况、表达水平及其和患者的预后关系。结果MTHFD2 mRNA在NMIBC组织内表达水平较正常组织高;NMIBCⅢ期、Ⅳ期组织内MTHFD2 mRNA表达高于Ⅱ期。82例患者中60例患者癌组织内MTHFD2蛋白表达为阳性,其阳性率为73.17%;其对应的癌旁组织内,存在14.63%表达为阳性。NMIBC组织内MTHFD2蛋白表达状况和肿瘤数目、大小、病理分期等存在关联(P<0.05)。MTHFD2蛋白阳性表达组患者术后无瘤生存时间较阴性表达组短(P<0.05)。结论NMIBC组织内MTHFD2水平的高表达提示患者可能存在预后不佳状况,其可作为NMIBC临床预后的潜在指标。
Objective To investigate the expression and significance of methylenetetrahydrofolate dehydrogenase 2(MTHFD2)in non muscle invasive bladder cancer(NMIBC).Methods Immunohistochemical staining was used to detect the expression of MTHFD2 in 82 patients with NMIBC.To analyze the expression of MTHFD mRNA in NMIBC tissues and its relationship with the prognosis of patients.Results The expression level of MTHFD2 mRNA in NMIBC was higher than that in normal tissues;The expression of MTHFD2 mRNA in NMIBC stageⅢandⅣtissues was higher than that in NMIBC stage II tissues.Among the 82 patients,60 patients showed positive expression of MTHFD2 protein in cancer tissues,with a positive rate of 73.17%.In the corresponding non cancerous tissues,14.63%of patients showed positive expression of MTHFD2 protein,and the expression of MTHFD2 protein in NMIBC tissues was correlated with the number,size and pathological stage of the tumor(P<0.05).The tumor free survival time of patients with positive expression of MTHFD2 protein was shorter than that of patients with negative expression of MTHFD2 protein(P<0.05).Conclusion The high expression of MTHFD2 in NMIBC suggests that patients may have a poor prognosis,which may be a potential indicator of clinical prognosis of NMIBC.
