摘要
目的探究沉默信息调节因子1(silent information regulator 1,SIRT1)、成纤维细胞活化蛋白-α(fibroblast activation protein-α,FAP-α)、半胱氨酸天冬氨酸蛋白酶3(cysteine aspartase-3,caspase-3)在结直肠癌中的表达及相关性。方法收集2021年4月至2022年4月整理归档较为完整的45例为结直肠癌组织石蜡标本,另外选取45例为距离癌组织边缘5 cm的癌旁组织石蜡标本。采取免疫组化法检测SIRT1、FAP-α、caspase-3水平表达,分析SIRT1、FAP-α、caspase-3在结直肠癌中的表达变化及与预后的相关性。结果与癌旁组织相比,结直肠癌组织中SIRT1、FAP-α、caspase-3表达量均有所升高,差异有统计学意义(P<0.05)。结直肠癌组织中SIRT1、FAP-α、caspase-3表达与结直肠癌患者的年龄、性别、肿瘤位置、远处转移无关,与淋巴结转移、肿瘤的直径、组织学类别、浸润程度、TNM分期、脉管瘤栓、分化程度有关相关,差异有统计学意义(P<0.05)。SIRT1、FAP-α、caspase-3之间Pearson相关性分析显示,SIRT1、FAP-α之间呈正相关(r=0.534,P=0.001);FAP-α、caspase-3之间呈正相关(r=0.556,P=0.001);SIRT1、caspase-3之间呈正相关(r=0.585,P=0.001)。ROC曲线显示,三项联合对结直肠癌的诊断价值高于SIRT1、FAP-α、caspase-3单项诊断(P=0.001)。结论结直肠癌患者机体内SIRT1、FAP-α、caspase-3水平表达上升,可看出其水平变化与结直肠癌患者产生病症有密切的关联,因此三者可作为预测结直肠癌患者治疗的重要依据。
Objective To explore the expression levels of silent information regulator 1(SIRT1),fibroblast activation protein-α(FAP-α),and cysteine aspartase-3(caspase-3)in colorectal cancer(CRC)and their correlation.Methods A total of 45 paraffin-embedded CRC tissues were collected from April 2021 to April 2022,and another 45 paraffin-embedded specimens surgically collected from 5cm away from the edge of cancer tissues were selected as controls.The expression levels of SIRT1,FAP-αand caspase-3 were determined by immunohistochemistry,and their correlation with the prognosis of CRC was analyzed.Results Compared with those of controls,SIRT1,FAP-α,and caspase-3 were significantly upregulated in CRC tissues(P<0.05).Their expressions in CRC tissues were not correlated with the age,sex,tumor location,and distant metastasis,but significantly correlated with lymph node metastasis,tumor diameter,histological category,infiltration degree,TNM staging,vascular tumor embolus,and differentiation degree(P<0.05).The Pearson correlation analysis showed a positive correlation between the expression levels of SIRT1 and FAP-α(r=0.534,P=0.001).A positive correlation between the expression levels of FAP-αand caspase-3 was identified(r=0.556,P=0.001),and meanwhile,a positive correlation was identified between the expression levels of SIRT1 and caspase-3(r=0.585,P=0.001).The ROC curve showed that the value of the combination detections of SIRT1,FAP-αand caspase-3 had a significantly higher diagnostic value in CRC than that of a single detection of either of them(P=0.001).Conclusion SIRT1,FAP-αand caspase-3 are upregulated in CRC patients,which are significantly correlated with the disease condition.The combination detection of them provides important references for predicting the prognosis of CRC.
