三七总皂苷与莪术醇联用对抗结肠癌细胞株HCT116和HT29的效果及机制研究 被引量:1

摘要 目的 探讨三七总皂苷、莪术醇联用对抗结肠癌细胞株HCT116和HT29的效果及其可能的机制。方法 采用体外培养HCT116及HT29结肠癌细胞系模型,分别给予三七总皂苷(50μmol/L)(三七总皂苷组)、莪术醇(50μmol/L)(莪术醇组)及联合用药(50μmol/L三七总皂苷+50μmol/L莪术醇)(联合组)进行处理,并将细胞培养24 h。以5-FU为阳性对照设置阳性对照组,同时设置空白对照组(只加细胞和培养液)。体外培养24、48 h后,统计细胞抑制率、细胞凋亡及细胞周期情况,采用免疫细胞化学法检测Ang-2和caspase-3蛋白的表达。结果 人结肠癌细胞经三七总皂苷、莪术醇以及联合处理后,细胞生长受到不同程度的抑制,联合组对HCT116和HT29的抑制率明显高于单药的应用,差异有统计学意义(P<0.05),且抑制程度随着时间的增加而增强;联合组干预48 h后人结肠癌HCT116和HT29细胞凋亡率明显高于各单用组,差异有统计学意义(P<0.05);联合组干预24 h后,HCT116、HT-29细胞发生变化,G1期细胞数增加,S期细胞数明显减少,差异有统计学意义(P<0.05);联合组干预24 h后,HCT116和HT29细胞中Ang-2的表达水平明显降低,caspase-3的表达水平则明显升高,差异有统计学意义(P<0.05)。结论 三七总皂苷、莪术醇联用能显著增强抗结肠癌作用,其机制可能是通过抑制细胞增殖,并促进细胞凋亡的双重作用。 Objective To investigate the effects and mechanism of Panax notoginseng saponins(PNS)and curcumenol against the colon cancer cell lines HCT116 and HT29.Methods HCT116 and HT29 colon cancer cell line models were cultured and treated with 50μmol/L PNS,50μmol/L curcumenol,and 50μmol/L PNS+50μmol/L curcumenol,respectively,for 24 h.A positive control(5-fluorouracil,5-FU)group and a blank control(only cells and medium)group were designed.After in vitro culture for 24 h and 48 h,the inhibition rate,apoptosis,and cell cycle were recorded,and immunocytochemistry was employed to measure the expression of angiotensin-2(Ang-2)and caspase-3.Results The treatment with PNS and/or curcumenol inhibited the growth of HCT116 and HT29 cells to different degrees,and the PNS+curcumenol group had higher inhibition rate than the PNS or curcumenol group(P<0.05).Furthermore,the inhibitory effect enhanced over time.The apoptosis rate of HCT116 and HT29 cells treated with PNS+curcumenol for 48 h was higher than that of the cells treated with PNS or curcumenol(P<0.05).The treatment with PNS+curcumenol for 24 h increased the cells in G1 phase and reduced the cells in S phase(P<0.05).Furthermore,the treatment with PNS+curcumenol for 24 h down-regulated the expression level of Ang-2 and up-regulated the expression level of caspase-3 in HCT116 and HT29 cells(P<0.05).Conclusion The combination of PNS and curcumenol can exert the anti-colon cancer effect by inhibiting cell proliferation and promoting apoptosis.
