基于数据挖掘和网络药理学探讨多系统萎缩的中医证治规律

摘要 目的:基于中医传承辅助平台及网络药理学,探讨多系统萎缩的中医证型分布、用药规律及核心药对作用机制。方法:检索中国知识资源总库、万方数据学术论文总库、维普中文科技期刊全文数据库,统计中医药治疗多系统萎缩的相关文献(建库以来至2022年6月),建立数据库;利用数据挖掘技术对用药进行挖掘分析;并利用网络药理学探讨核心药对的作用机制。结果:本研究共筛选出关于中医药治疗多系统萎缩的文献62篇,含处方82首,涉及中药162味,得到中医证型、高频用药及其相对应的性味、归经。关联规则分析得出药对15对,聚类分析得到3类核心药物组合,发现核心药对为熟地黄-山茱萸;活性成分与疾病交集靶点有55个;药对活性成分主要通过神经活性配体受体相互作用、癌症通路、钙信号通路等发挥作用。结论:现代中医辨证多系统萎缩以虚证为主;核心治法为温补脾肾,滋补肝肾;核心药对为熟地黄-山茱萸;药对活性成分主要通过神经活性配体受体相互作用、癌症通路、钙离子信号通路等调控多系统萎缩。 Objective:To explore the distribution of TCM patterns,medication rules and the mechanism of core medicine pairs of multiple system atrophy based on the Traditional Chinese Medicine Inheritance Support Platform and network pharmacology methods.Methods:China National Knowledge Infrastructure database,Wanfang database and VIP database for Chinese technical journals were used to search relevant literature on the treatment of multiple system atrophy in TCM from the time of construction to June 2022,a database was established,and the data mining technique was used to perform a statistical analysis for TCM medicines,and network pharmacology method was used to analyze the mechanism of action of the core drug pairs.Results:A total of 62 studies on treating multiple system atrophy were retrieved and screened,containing 82 prescriptions involving a total of 162 TCM medicines.The TCM syndrome types,high-frequency drugs and their corresponding properties,tastes and meridians were obtained.The association rules yielded 15 pairs of drugs,and the cluster analysis yielded 3 categories of core medicine combinations.The Shudihuang(Radix Rehmanniae Preparata)-Shanzhuyu(Fructus Corni)drug pair was the core drug,active ingredients and disease intersected 55 targets.The active ingredients of drug pairs exert their effects mainly through neuroactive ligand-receptor interaction,pathways in cancer,calcium signaling pathway and so on.Conclusion:The analysis reflects that the typical syndromes of multiple system atrophy are based on deficiency syndrome in modern TCM,and the main treatment method was warming spleen and kidney,invigorating liver and kidney.Shudihuang-Shanzhuyu drug pair was the core drug pair.The active ingredients of drug pair regulate multiple system atrophy through neuroactive ligand-receptor interaction,pathways in cancer,calcium signaling pathway and so on.
