摘要
脑卒中的发病机制是一个动态演变过程,表现为一系列神经毒性和神经保护相互作用的反应,参与反应的蛋白质作为信号通路通常在外界刺激下激活。目前已明确针刺能通过抑制脑卒中后脑组织神经细胞铁死亡从而促进神经功能修复,近年来针刺通过调控铁死亡治疗脑卒中的相关信号通路研究取得了显著进展,主要包括以下几方面:miR-23a-3p和核转录因子红系2样2(NFE2L2)信号通路;p62/Kelch样ECH关联蛋白1/核因子E2相关因子2(p62/Keap1/Nrf2)信号通路;α7nAChR介导的Janus激酶2/信号转导及转录激活因子3(JAK2/STAT3)信号通路;环磷酸腺苷/cAMP依赖性蛋白激酶/cAMP反应元件结合蛋白(cAMP/PKA/CREB)信号通路;磷脂酰肌醇-3激酶/蛋白激酶B(PI3K/AKT)信号通路;p38丝裂原活化蛋白激酶(MAPK)信号通路;过氧化物酶体增殖物激活受体γ辅激活因子1α/线粒体转录因子A(PGC-1α/TFAM)信号通路;Notch信号通路和脑源性神经营养因子(BDNF)介导的Nrf2信号通路。其中有已被证实的信号通路,亦有待研究的信号通路。
The pathogenesis of stroke is a dynamic evolution process with the interactions between neurotoxicity and neuroprotection,and the proteins as signaling pathways involved in the interactions are usually activated by external stimuli.Studies have proved that acupuncture can inhibit ferroptosis of nerve cells in the brain after stroke to promote restoration of the neural function.In recent years,progress has been achieved in the regulation of ferroptosis-related signaling pathways by acupuncture after cerebral stroke.These pathways involve miR-23a-3p/erythroid-derived 2-like protein 2(NFE2L2),p62/Kelch-like ECH-assocated protein 1(Keap1)/nuclear factor erythroid 2-related factor 2(Nrf2);α7nAChR-mediated Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3);cyclic adenosine monophosphate(cAMP)/cAMP-dependent protein kinase(PKA)/cAMP response element binding protein(CREB),phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT),p38 mitogen-activated protein kinase(MAPK),peroxisome proliferator-activated receptor-gamma coactivator-1 alpha(PGC-1α)/TFAM,and Notch and brain-derived neurotrophic factor(BDNF)-mediated Nrf2.Some of these signaling pathways have already been confirmed and others remain to be studied.
