摘要
目的:探讨川乌与草乌中生物碱类化学成分的差异。方法:采用超高效液相色谱-四极杆-静电场轨道阱串联质谱(UHPLC-Q-Exactive Orbitrap MS/MS)比较分析川乌与草乌中的生物碱类成分的差异;采用超声波提取联合强阳离子吸附树脂固相萃取分离的方法富集纯化川乌和草乌中的生物碱类成分;利用ACQUITY BEH C18 column色谱柱,以0.1%氨水-乙腈为流动相进行梯度洗脱,ESI正离子模式下采集对照品与样品的高分辨质谱数据。通过与对照品的保留时间及质谱数据信息对比,并结合文献报道对川乌、草乌中的生物碱类成分进行比较分析。结果:从川乌中分析鉴定了178个成分,从草乌中鉴定了171个成分,其中两者共有成分164个,川乌特有成分14个,草乌特有成分7个。草乌中主要双酯型生物碱成分的含量高于川乌。结论:川乌和草乌生物碱类成分谱基本一致,是两者性味、药性相似的基础。双酯型生物碱成分含量高可能是导致草乌药效及毒性强于川乌的原因。
Objective:To explore the differences in the chemical constituents of alkaloids in Aconiti Radix and Aconiti Kusnezoffii Radix.Methods:Ultra-Performance liquidchromatography-Quadrupole-electrostatic field Orbitrap mass spectrometry(UHPLC-Q-Exactive Orbitrap MS/MS)was used to compare the differences of the alkaloids in Aconiti Radix and Aconiti Kusnezoffii Radix.The alkaloids in Aconiti Radix and Aconiti Kusnezoffii Radix were enriched and purified by ultrasonic extraction combined with strong cation adsorption resin solid phase extraction.The gradient elution was performed on ACQUITYTM BEH C18 column with 0.1%ammonia water-acetonitrile as mobile phase.High-resolution mass spectrometry data of controls and samples collected in ESI positive ion mode.The alkaloids in Aconiti Radix and Aconiti Kusnezoffii Radix were compared and analyzed by comparing with the retention time and mass spectrometry data of the reference substance and combining with the literature reports.Results:A total of 178 components were identified in Aconiti Radix and 171 components were identified in Aconiti Kusnezoffii Radix with 164 components in common.A total of 14 specific components were detected in Aconiti Radix.And 7 specific components were detected in Aconiti Kusnezoffii Radix.The content of the main diester alkaloids in Aconiti Kusnezoffii Radix was higher than that of Aconiti Radix.Conclusion:The composition spectrum of alkaloids in Aconiti Radix and Aconiti Kusnezoffii Radix are in substantial agreement,which is the basis for the similar nature and flavor and medicinal properties of each other.The content of the main diester alkaloids in Aconiti Kusnezoffii Radix is higher than that of Aconiti Radix,which may be the reason that the efficacy and toxicity of Aconiti Kusnezoffii Radix are stronger than those of Aconiti Radix.
