摘要
目的 分析叶酸代谢相关酶基因多态性与不良出生结局发生易感性的相关性。方法 选取2020年1月至2022年12月临夏回族自治州人民医院收取的3 467名产妇作为研究对象。根据出生结局分为正常组(n=2 182)与不良结局组(n=1 285),通过聚合酶链反应(PCR)-荧光探针法检测亚甲基四氢叶酸还原酶(MTHFR)与甲硫氨酸合成酶还原酶(MTRR)基因多态性,使用Logistic回归模型分析基因交互作用。结果 不良结局组MTHFR C677T TT基因型频率与T等位基因频率显著高于正常组(P<0.05);不良结局组MTHFR A1298C CC基因型频率与C等位基因频率显著高于正常组(P<0.05);不良结局组MTRR A66G GG基因型频率与G等位基因频率显著高于正常组(P<0.05);Logistic回归分析显示,MTHFR A1298C和MTRR A66G具有交互作用,能够促使不良出生结局危险性变大(OR=1.614,P<0.001)。结论 MTHFR C677T、A1298C位点及MTRR A66G位点与不良出生结局密切相关,MTHFR A1298C和MTRR A66G交互作用能够增加不良出生结局发生易感性。
Objective To analyze the correlation between folic acid metabolism-related enzyme gene polymorphism and susceptibility to adverse birth outcomes.Methods A total of 3,467 parturients from the People′s Hospital of Linxia were selected as research subjects and divided into the normal group(n=2,182)and the adverse outcome group(n=1,285)according to birth outcomes.Gene polymorphisms of methylenetetrahydrofolate reductase(MTHFR)and methionine synthase reductase(MTRR)were monitored through polymerase chain reaction(PCR)-fluorescence probe method.Genetic interactions were analyzed by Logistic regression model.Results TT genotype frequency and T allele frequency of MTHFR C677T in the adverse outcome group were significantly higher than those in the normal group(P<0.05).CC genotype frequency and C allele frequency of MTHFR A1298C in the adverse outcome group were significantly higher than those in the normal group(P<0.05).GG genotype frequency and G allele frequency of MTRR A66G in the adverse outcome group were significantly higher than those in the normal group(P<0.05).Logistic regression analysis showed that there was an interaction between MTHFR A1298C and MTRR A66G,which could increase the risk of adverse birth outcomes(OR=1.614,P<0.001).Conclusions MTHFR C677T and A1298C,and MTRR A66G loci are closely related to adverse birth outcomes.The interaction between MTHFR A1298C and MTRR A66G can increase the susceptibility to adverse birth outcomes.
