青果不同浓度乙醇提取物对营养性肥胖模型大鼠减肥作用的影响 被引量:1

Effects of Different Concentrations of Ethanol Extract of Qingguo(Canarii Fructus)on Weight Loss in Nutritive Obesity Model Rats

摘要 目的:研究青果不同浓度乙醇提取物对营养性肥胖模型大鼠体质量增长的影响,并探索青果发挥减肥作用的机制。方法:将70只SPF级雄性SD大鼠随机分为普食组、模型组、奥利司他组及青果水提取组、50%乙醇提取组、70%乙醇提取组、95%乙醇提取组,每组各10只,适应性饲养7 d后,各组分别灌胃给药(普食组、模型组灌胃生理盐水;各提取组灌胃对应青果提取物,给药剂量为3.0 g/kg;奥利司他组灌胃奥利司他溶液,给药剂量为10.8 mg/kg),连续灌胃30 d。模型组和各给药组普通饲料喂养10 d后,改为高脂饲料喂养20 d。观察不同时期大鼠体征、体质量、Lee’s指数、粪便总脂含量、血清血脂指标、小肠α-葡萄糖苷酶活性、腹部及附睾脂肪率、脂肪组织形态变化。结果:灌胃20 d时,模型组大鼠体质量增长明显高于普食组(P<0.01),说明造模成功。灌胃30 d时,各青果提取组体质量增长低于模型组(P<0.01),以50%乙醇提取组和70%乙醇提取组下降最为明显。与模型组比较,各给药组大鼠粪便总脂含量均升高(P<0.01);青果各乙醇提取组大鼠Lee’s指数均降低(P<0.05),以70%乙醇提取组下降最明显;青果各乙醇提取组大鼠高密度脂蛋白胆固醇含量升高,低密度脂蛋白胆固醇、甘油三酯、总胆固醇含量均降低(P<0.01),其中50%乙醇提取组和70%乙醇提取组血脂指标变化最明显;青果各乙醇提取组腹部脂肪率显著降低(P<0.05),70%乙醇提取组附睾脂肪率显著降低(P<0.05);各青果提取组大鼠小肠α-葡萄糖苷酶活性均下降(P<0.05或P<0.01),以70%乙醇提取物组下降最为明显。模型组附睾组织细胞数量明显减少,细胞边界不清晰,细胞形状不规则,含有大量脂滴,有明显炎症细胞浸润现象。各给药组附睾组织细胞体积显著变小,排列致密,细胞边界清晰,含有少量脂滴或无脂滴,有较轻炎症或无炎症细胞浸润现象,其中以青果50%乙醇提取组和70%乙醇提取组表现最佳。结论:青果不同浓度乙醇提取物可以通过抑制α-葡萄糖苷酶活性发挥降血脂作用,防止脂肪沉积和体质量增长;青果不同浓度乙醇提取物均具有预防肥胖、降脂减肥作用,以70%乙醇提取物作用效果最佳。 Objective:To study the effects of different concentrations of ethanol extract of Qingguo(Canarii Fructus)on weight gain of nutritive obesity model rats,and to explore the mechanism of Qingguo on weight loss.Methods:Total 70 SPF male SD rats were randomly divided into general diet group,model group,orlistat group,and Qingguo water extraction group,50%ethanol extraction group,70%ethanol extraction group,95%ethanol extraction group,with 10 rats in each group.After 7 days of adaptive feeding,each group was given intragastric administration(General diet group and model group were given normal saline;Intragastric admini-stration of the corresponding Qingguo extract in each extraction group was 3.0 g/kg;Orlistat group was given orlistat solution at the dose of 10.8 mg/kg)for 30 days.The model group and each administration group were fed with ordinary diet for 10 days,and then fed with high-fat diet for 20 days.The changes of physical signs,body mass,Lee’s index,total fat content in feces,serum lipid index,small intestineα-glucosidase activity,abdominal and epididymal fat rate,adipose tissue morphology of rats were investigated in different periods.Results:After 20 days of intragastric administration,the increase of body mass in model group was significantly higher than that in general diet group(P<0.01),indicating that the molding is successful.After 30 days of intragastric administration,the growth of body mass of each Qingguo extraction group were lower than that of the model group(P<0.01),and the decrease of 50%ethanol extraction group and 70%ethanol extraction group was most significant.Compared with model group,total fat content in feces of rats in all administration groups was increased(P<0.01);Lee’s index decreased in all Qingguo ethanol extraction groups(P<0.05),and the decrease was most significant in 70%ethanol extraction group;The content of high-density lipoprotein cholesterol increased,and the content of low-density lipoprotein cholesterol,triglycerides,and total cholesterol decreased in all Qingguo ethanol extraction groups(P<0.01),among which 50%ethanol extraction group and 70%ethanol extraction group showed the most significant changes in blood lipid indexes;The percentage of abdominal fat in all Qingguo ethanol extraction group was significantly decreased(P<0.05),and the fat percentage of epididymis in 70%ethanol extraction group was significantly decreased(P<0.05);The activity ofα-glucosidase in small intestine of rats in all Qingguo extraction groups decreased(P<0.05 or P<0.01),and the decrease was most significant in 70%ethanol extract group.In the model group,the number of cells in epididymal tissue was significantly reduced,the cell boundary was not clear,and the cell shape was irregular,there were a lot of fat drops,and there was obvious inflammatory cell infiltration.The cell volume of epididymal tissue in each administration group was significantly smaller,the arrangement was dense,and the cell boundary was clear,there were few fat drops or no fat drops,and there was light inflammatory or no inflammatory cell infiltration,and the 50%ethanol extraction group and 70%ethanol extraction group performed the best.Conclusions:The ethanol extracts of different concentrations of Qingguo play a role in lowering blood lipid,preventing fat deposition and body mass growth by inhibitingα-glucosidase activity.Different concentrations of ethanol extract of Qingguo can prevent obesity,reduce fat and lose weight.The effect of 70%ethanol extractive was the best.
