热带生态区玉米自交系离体花粉活性的变化特征

Variation Characteristics of in Vitro Pollen Activity of Maize Inbred Lines Planted in Tropical Ecological Area

摘要 2020、2021年在海南热带生态区以6个不同基因型玉米自交系ZM4321、驻136、驻54、ZM3358、H1269、ZM027为材料,离体花粉在4℃冷藏、‒7℃冷冻、室温条件下贮存一定时间,授粉后通过田间结实率,研究了Reid类和Lancaster类自交系离体花粉不同温度贮存条件下其活性持续时间,通过TTC染色率鉴定离体花粉的活性。结果表明:不同温度贮存条件下,自交系离体花粉的活性随贮存时间的增加而下降,而离体花粉活性衰减变化在自交系间存在差异。4℃贮存条件下,ZM4321、H1269离体花粉活性持续长达54 h,而驻136在10 h后结实率急剧降低,驻54、ZM3358花粉活性主要集中于8 h内,ZM027贮存48 h结实率约20%。‒7℃贮存1 h时,ZM3358结实率超85%,2 h时结实率2020、2021年分别下降至2.64%、1.18%;ZM3358离体花粉对‒7℃贮存极度敏感,而WS2-4321、H1269、ZM027花粉高结实率持续时间最长,为12 h,明显高于驻136、驻54。室温贮存条件下,ZM4321、H1269离体花粉高结实率的维持时间高达30 h;ZM3358、ZM027离体花粉高结实率维持时间为10~11 h,但11 h后ZM027结实率下降幅度远大于ZM3358;驻136离体花粉高结实率维持时间为11 h;驻54离体花粉高结实率维持不超过3 h。ZM4321在4℃贮存,驻136、驻54在‒7℃贮存,ZM3358在室温贮存更有利于离体花粉活性的保持,而H1269、ZM027在3种温度贮存条件下离体花粉活性差异不大。离体花粉TTC染色率与田间结实率表现一致。 In 2020 and 2021,six different genotypes of maize inbred lines ZM4321,Zhu136,Zhu 54,ZM3358,H1269,and ZM027,were used as materials in a Hainan tropical ecological area.The in vitro pollen was stored at 4,7℃,and room tem-perature for some time.After pollination,in vitro pollen activity of Reid type and Lancaster type inbred lines under different temperature storage conditions was studied through field seed setting rates.The in vitro pollen activity was identified by TTC staining rate.The results showed that in vitro pollen activity of inbred lines under different temperature storage conditions decreased with the delay of storage time increasing,and the attenuation of in vitro pollen activity was varied among inbred lines.The in vitro pollen activity of ZM4321 and H1269 was stored at 4℃lasted for 54 h,and the seed setting rate of Zhu136 decreased sharply after 10 hours,the pollen activities of Zhu54 and ZM3358 were mainly concentrated within 8 hours,and the seed setting rate of ZM027 was about 20%after 48 hours.When stored at‒7℃for 1 h,the seed setting rate of ZM3358 ex-ceeded 85%,and the seed setting rate in 2 h decreased to 2.64%and 1.18%in 2020 and 2021,respectively.ZM3358 in vitro pollen was extremely sensitive to low temperature,while the seed setting rate of ZM4321,H1269,and ZM027 pollen was the longest for 12 h,significantly higher than Zhu136 and Zhu54.Under the condition of room temperature storage,the mainte-nance time of seed setting rate of ZM4321 and H1269 in vitro pollen was as high as 30 h,the maintenance time of high seed setting rate of ZM3358 and ZM027 in vitro pollen was 10-11 h,after 11 h,the decline range of ZM027 seed setting rate was much higher than ZM3358,the maintenance time of high seed setting rate of Zhu136 in vitro pollen was 11 h,the high seed setting rate of Zhu54 in vitro pollen was maintained for no more than 3 h.ZM4321 stored at 4℃,Zhu136 and Zhu54 stored at‒7℃,and ZM3358 stored at room temperature were more conducive to the in vitro pollen activity maintenance.However,H1269 and ZM027 have little difference under the three temperature storage conditions.The TTC staining rate of in vitro pol-len was consistent with the field seed setting rate.
