摘要
目的探讨藿苓益智方通过调控MAPK信号通路对PC12细胞帕金森病模型凋亡、炎性因子和氧化应激的影响。方法体外培养PC12细胞,用1-甲基-4-苯基吡啶离子(1-methyl-4-phenylpyridinium,MPP+)诱导建立帕金森病模型,将细胞分为对照组、模型组、藿苓益智方低剂量组、藿苓益智方高剂量组、藿苓益智方+Anisomycin组。用CCK8法检测细胞增殖活性;流式细胞术检测细胞凋亡;Western blot检测Bax、Bcl-2、p38、p-p38、p-JNK、JNK蛋白表达;ELISA法检测SOD活性、GSH-Px活性、TNF-α含量、IL-6含量。结果与对照组比较,模型组细胞活性、Bcl-2蛋白表达显著减少(P<0.05)、SOD活性、GSH-Px活性明显降低(P<0.05),凋亡率、Bax、p-p38、p-JNK蛋白表达、TNF-α含量、IL-6含量明显增加(P<0.05)。与模型组比较,藿苓益智方低剂量组、藿苓益智方高剂量组细胞活性、Bcl-2蛋白表达、SOD活性、GSH-Px活性明显增加(P<0.05),凋亡率、Bax、p-p38、p-JNK蛋白表达、TNF-α含量、IL-6含量明显降低(P<0.05)。与藿苓益智方组比较,藿苓益智方+Anisomycin组细胞活性、Bcl-2蛋白表达、SOD活性、GSH-Px活性明显降低(P<0.05),凋亡率、Bax蛋白表达、TNF-α含量、IL-6含量明显增加(P<0.05)。结论藿苓益智方通过激活MAPK信号通路促进MPP+诱导PC12细胞凋亡、氧化应激和炎性损伤。
Objective To explore the effects of Huoling Yizhi recipe on apoptosis,inflammatory factors,and oxidative stress of PC12 cells in the Parkinson's disease model by regulating the MAPK signal pathway.Methods PC12 cells were cultured in vitro and induced with 1-methyl-4-phenylpyridinium(MPP+)to establish a Parkinson's disease model.The cells were divided into the control group,model group,low-dose Huoling Yizhi recipe group,high-dose Huoling Yizhi recipe group,and Huoling Yizhi recipe+Anisomycin group.CCK8 method was adopted to detect cell proliferation activity;flow cytometry was used to detect cell apoptosis;Western blot was utilized to detect Bax,Bcl-2,p38,p-p38,p-JNK,and JNK protein expression;ELISA method was employed to detect SOD activity,GSH-Px activity,TNF-αcontent,and IL-6 content.Results Compared with the control group,the cell activity,Bcl-2 protein expression,SOD activity,and GSH-Px activity of the model group were significantly reduced(P<0.05),and the apoptosis rate,Bax,p-p38,p-JNK protein expression,TNF-αcontent,and IL-6 content of the model group were significantly increased(P<0.05).Compared with the model group,the cell activity,Bcl-2 protein expression,SOD activity,and GSH-Px activity in the low-dose and high-dose Huoling Yizhi recipe groups increased significantly(P<0.05),and the apoptosis rate,Bax,p-p38,P-JNK protein expression,TNF-αcontent,and IL-6 content were significantly reduced(P<0.05).Compared with the Huoling Yizhi recipe group,the cell activity,Bcl-2 protein expression,SOD activity,and GSH-Px activity in the Huoling Yizhi recipe+Anisomycin group were significantly reduced(P<0.05),and the apoptosis rate,Bax protein expression,TNF-αcontent,and IL-6 content significantly increased(P<0.05).Conclusion Huoling Yizhi recipe promotes MPP+induced PC12 cell apoptosis,oxidative stress,and inflammatory damage by activating the MAPK signaling pathway.
