红豆杉根际益生菌的筛选及其抗病促生作用研究 被引量:1

Screening of Probiotics from Taxus Rhizosphere and Its Effect on Disease Resistance and Growth Promotion

摘要 炭疽病是红豆杉幼苗普遍发生的病害之一,建立其绿色生物防治技术具有重要意义。本文从健康红豆杉根际土壤中分离具有抗病促生作用的益生菌,采用平板对峙试验以及盆栽防病试验评估其拮抗能力,通过溶磷、产IAA和分泌嗜铁素等试验以及红豆杉枝条扦插生根试验探究其对红豆杉的促生作用。结果表明,筛选出4株对红豆杉炭疽病病原菌具有明显防治效果的菌株MK17(Burkholderia stabilis)、MK18(Paraburkholderia caffeinilytica)、MK30和MK128(Burkholderia pyrrocinia),其中MK30和MK128抑菌率均为56.86%。盆栽防病试验表明,4株拮抗菌的防治效果均达到30%以上,且MK128菌株在加入阿拉伯胶、吐温-20和抗坏血酸混合液后,防治率提升到63.24%,达到了与化学药物苯醚甲环唑相同的效果。同时,这4株菌均至少具有溶磷、产IAA和分泌嗜铁素中的一种活性,其中MK18分泌IAA可达21.03±0.20 mg/L,MK128溶磷和分泌嗜铁素D/d值分别为6.16±0.09 mg/L和2.45±0.03。扦插试验表明,与生根粉相比,MK18和MK128单独或联合处理均能显著促进红豆杉枝条生根,且两个菌联合处理的效果优于单菌处理。上述结果为红豆杉炭疽病的绿色生物防治提供了依据和优良的益生菌株。 Anthracnose is one of the common diseases of Taxus seedlings,and it is of great significance to establish its green biological control technology.In this study,the probiotics with disease resistance and growth promotion effects were isolated from the rhizosphere soil of healthy Taxus chinensis.The antagonistic ability of the probiotics was evaluated by the plate confrontation test and potted disease control experiments,and the growth promotion effect was explored through experiments such as phosphorus dissolution,IAA production and iron carrier production,as well as the rooting test of T.chinensis cuttings.The results showed that four strains MK17(Burkholderia stabilis),MK18(Paraburkholderia caffeinilytica),MK30(Burkholderia pyrrocinia)and MK128(Burkholderia pyrrocinia)had obvious control effects on the pathogen of anthracnose of T.chinensis by screening plate confrontation method,and the inhibition rate of MK30 and MK128 reached 56.86%.Potted disease control experiments showed that the control effects of the four antagonistic bacteria all reached more than 30%,and the control rate of MK128 increased to 63.24%after adding a mixture of gum arabic,Tween-20 and ascorbic acid,reaching the same control effect of difenoconazole.At the same time,the four antagonistic bacteria all had at least one of the activities of phosphorus dissolution,IAA production or iron carrier production,among which MK18 could secrete IAA up to 21.03±0.20 mg/L,and MK128 could dissolve Phosphorus and secrete Siderophore with D/d values of 6.16±0.09 mg/L and 2.45±0.03,respectively.The cutting experiment showed that MK18 and MK128 individually or combined treatment could significantly promote the rooting of T.chinensis branches comparing with the rooting powder,and the effect of combined treatment of two bacteria was better than that of single bacteria treatment.The above results provide experimental basis and excellent probiotic strains for the biological control of Taxus anthracnose.
