白藜芦醇对低压低氧诱导大鼠牙槽骨损伤的干预作用研究 被引量:1

INTERVENTION 0F HYPOBARIC HYPOXIA-INDUCED ALVEOLAR BONE INJURY BY RESVERATROL IN RATS

摘要 目的研究低气压缺氧环境对牙槽骨的损伤及白藜芦醇抗牙槽骨骨质疏松的效果。方法雄性Wistar大鼠36只随机分为常氧组(control)、低压低氧组(hypoxia)和白黎芦醇干预组(Res),每组12只。采用低压氧舱模拟高原低压低氧环境,制备大鼠高原低氧模型,白藜芦醇干预组使用400mg/kg白藜芦醇进行灌胃,常氧组和低压低氧组使用0.5%羧甲基纤维素钠作为对照。各组处理9 w后进行动物安乐死并采集骨样本,通过Micro-CT、组织学染色及免疫组织化学染色分析,观察牙槽骨骨组织显微结构及成骨破骨分化情况。结果Micro-CT及HE染色显示:与常氧组相比,低压低氧组大鼠下颌第一磨牙根分歧下方骨小梁变少且稀疏,牙根之间骨质破坏严重,BV/TV和Tb.N降低,Tb.Sp升高,差异均具有显著统计学意义(P<0.01);与低压低氧组相比,白藜芦醇干预组大鼠骨小梁变化均趋于正常,BV/TV和Tb.N升高,Tb.Sp降低,差异具有统计学意义(P<0.05)。与常氧组相比,白藜芦醇干预组大鼠BV/TV和Tb.N降低,差异具有统计学意义(P<0.05);Tb.Sp差异无统计学意义(P>0.05)。组织特殊染色及免疫组化染色显示:与常氧组相比,低压低氧组TRAP、HIF-1α阳性染色区域显著增加(P<0.01),OCN阳性染色区域减少(P<0.05),Runx2、PHD2阳性染色区域显著减少(P<0.01);与低压低氧组相比,白藜芦醇干预组TRAP、HIF-1α阳性染色区域显著减少(P<0.01),OCN阳性染色区域增加(P<0.05),而趋近于常氧组,差异无统计学意义(P>0.05),Runx2阳性染色区域显著增加(P<0.01),而较低于常氧组(P<0.05),PHD2阳性染色区域显著增加(P<0.01),而显著低于常氧组(P<0.01)。结论本研究初步验证了低氧低压环境可能导致大鼠牙槽骨产生与股骨类似的骨代谢紊乱,继而可能引发颌骨骨质疏松等相关疾病,对于口腔种植及牙周疾病等牙槽骨缺损相关治疗有所影响。白藜芦醇通过促进骨形成及减缓骨吸收,可有效对抗高原缺氧环境诱导的下颌骨骨质疏松,潜在机制尚需进一步研究。 Objective To explore the damage of alveolar bone induced by hypobaric hypoxia and the intervention by resveratrol.Methods Thirty-six male Wistar rats were randomly divided into normoxia group(control),hypoxia group(Hypoxia)and resveratrol group(Res),with 12 rats in each group.A hypobaric oxygen chamber was used to simulate the plateau hypobaric and hypoxic environment.The resveratrol group was given 400 mg/kg resveratrol by gavage,and the normoxia and hypoxia groups were given 0.5%sodium carboxymethyl cellulose as a control.Animals were euthanized and bone samples were collected after 9 weeks of treatment.Micro-CT,histological staining and immunohistochemical staining were used to analyze the microstructure of bone tissue and osteoclast differentiation.Results Micro-CT and HE staining showed that compared with the normoxia group,the trabecular bone below the bifurcation of the mandibular first molar roots in the hypoxia group was less and sparse,and the bone between the roots severely damaged,and BV/TV and Tb.N decreased.Compared with the hypoxia group,the changes of trabecular bone in the resveratrol group tended to be normal.BV/TV and Tb.N were increased,and Tb.Sp decreased(P<0.05).Compared with the normoxia group,the BV/TV and Tb.N of the resveratrol group were decreased,and the difference was statistically significant(P<0.05).There was no significant difference in Tb.Sp(P>0.05).Tissue special staining and immunohistochemical staining showed that compared with the normoxia group,the positive staining areas of TRAP and HIF-1αin the hypoxia group were significantly increased(P<0.01),and the positive staining areas of OCN decreased(P<0.05).The positive staining area of Runx2,PHD2 was significantly reduced(P<0.01).Compared with the hypoxia group,the TRAP and HIF-lαpositive staining areas in the resveratrol group were significantly reduced(P<0.01),and the OCN positive staining area increased(P<0.05).The Runx2 positive staining area was significantly increased(P<0.01),but lower than the normoxia group(P<0.05).The PHD2 positive staining area was significantly increased(P<0.01),but significantly lower than the normoxia group(P<0.01).Conclusion Hypoxic and low pressure environment may cause osteoporosis and other related diseases in the alveolar bone similar to the bone metabolism disorder in the femur.Resveratrol can effectively protect against mandibular osteoporosis induced by high altitude hypoxia by promoting bone formation and slowing down bone resorption.The potential mechanism needs further study.
