摘要
目的探讨三七总皂苷(PNS)对重症急性胰腺炎(SAP)病理、炎症、感染、肝功能和心肌酶谱的影响。方法SD大鼠随机分为Sham(假手术组)、SAP(模型组)、L-PNS、H-PNS和SP600125组;L-PNS、H-PNS和SP600125组分别在造模前腹腔注射低剂量PNS,高剂量PNS和阳性药物SP600125。造模后3、6、12和24 h,分别取胰腺和肺组织进行苏木精-伊红染色,取肺组织进行免疫组化染色和MPO,IL-1β和TNF-α水平的检测,取血清进行CRP,PCT,IL-6,α-AMS,ALT,CK-MB和LDH水平的检测。结果SAP模型构建成功,且相较于各时间点的SAP组,对应各时间点的L-PNS,H-PNS和SP600125组中胰腺组织和肺组织病理评分、肺组织中ICAM-1阳性细胞比例及MPO,IL-1β和TNF-α水平、血清中CRP,PCT,IL-6,α-AMS,ALT,CK-MB和LDH水平均显著降低(P<0.05)。这些指标在各时间点的H-PNS和SP600125组中无显著性差异(P>0.05)。结论PNS有效缓解胰腺损伤、肺损伤、肝功能和心肌酶谱异常。
Objective To investigate the effects of Panax Notoginseng Saponin(PNS)on the pathology,inflammation,infection,liver function and myocardial enzyme of severe acute pancreatitis(SAP).Method SD rats were randomly divided into Sham(sham-operated group),SAP(model group),L-PNS,H-PNS and SP600125 groups;L-PNS,H-PNS and SP600125 groups were injected intraperitoneally with low-dose PNS,high-dose PNS and positive drug SP600125 before molding.At 3h,6h,12h and 24h after molding,pancreas and lung tissues were taken for hematoxylin-eosin staining,lung tissues were taken for immunohistochemical staining and measurement of MPO,IL-1β and TNF-α levels,and serum was taken for measurement of CRP,PCT,IL-6,α-AMS,ALT,CK-MB and LDH levels.Results The SAP model was successfully constructed.Compared to the SAP group at each time point,the pancreatic tissue and lung histopathological scores,the proportion of ICAM-1 positive cells and the levels of MPO,IL-1β and TNF-α in lung tissue,as well as the levels of CRP,PCT,IL-6,α-AMS,ALT,CK-MB and LDH in serum were significantly reduced(P<0.05).These indicators were not significantly different between the H-PNS and SP600125 groups at each time point(P>0.05).Conclusion PNS effectively alleviates pancreatic and lung injury,as well as abnormal liver function and myocardial enzyme,providing potential application to clinical treatment strategies for SAP.
