龟板固本方对肺癌骨髓抑制小鼠骨髓细胞周期及凋亡相关蛋白表达的影响

摘要 目的研究龟板固本方对肺癌骨髓抑制小鼠骨髓细胞DNA周期和凋亡相关蛋白的影响,探讨其促进骨髓抑制肺癌小鼠造血功能恢复的分子机制。方法将60只C57BL/6小鼠随机分为空白组、模型组、龟板固本方组、粒细胞刺激因子组,每组15只。除空白组外,其他3组接种Lewis肺癌细胞建立肺癌模型,然后腹腔注射环磷酰胺制备肺癌骨髓抑制模型。造模完成后24 h开始给药,龟板固本方组给予龟板固本方悬液0.4 mL灌胃,空白组和模型组给等量生理盐水灌胃,粒细胞刺激因子组皮下注射重组人粒细胞刺激因子注射液5 ng/g,均1次/d,连续7 d。处死小鼠,采用流式细胞仪检测骨髓细胞周期,免疫组化法检测细胞凋亡相关蛋白Bcl-2和Bax表达情况。结果与模型组比较,龟板固本方组和粒细胞刺激因子组G_(0)/G_(1)期细胞比例和骨髓细胞中Bax蛋白表达量均明显降低(P均<0.05),S期细胞比例、G_(2)/M期细胞比例和骨髓细胞中Bcl-2蛋白表达量均明显升高(P均<0.05);与粒细胞刺激因子组比较,龟板固本方组S期细胞比例和骨髓细胞中Bcl-2蛋白表达量均较低(P均<0.05)。结论龟板固本方可通过上调骨髓细胞中Bcl-2蛋白表达和下调Bax蛋白表达,抑制骨髓细胞凋亡,使骨髓细胞进入S期和G_(2)/M增殖周期,促进骨髓细胞增殖,从而改善骨髓抑制。 Objective Ii is to investigate the effects of tortoise-plastron decoction for enhancing the root(TDER)on DNA cycle of bone marrow cells and apoptosis-related proteins in mice with lung cancer and myelosuppression,and to explore the molecular mechanism of this decoction in promoting the recovery of hematopoietic function of the mice.Methods Sixty C57BL/6 mice were randomly divided into blank group,model group,TDER group,and granulocyte stimulating factor group,with 15 mice in each group.Except for the blank group,the other three groups were inoculated with Lewis lung cancer cells to establish models of lung cancer,and then given cyclophosphamide by intraperitoneal injection to prepare models of lung cancer with myelosuppression.24 hours after the completion of modeling,the administration of drugs was started.The TDER group was given TDER suspension 0.4 mL by gavage,the blank group and model group were given an equal amount of normal saline gavage,and the granulocyte stimulating factor group was injected subcutaneously with recombinant human granulocyte-stimulating factor injection 5 ng/g,all once daily,continuously treated for 7 days.The mice were executed,and the bone marrow cell cycle was detected by flow cytometry,the expression of apoptosis-related proteins Bcl-2 and Bax was detected by immunohistochemistry.Results Compared with the model group,the proportion of G_(0)/G_(1)-phase cells and the expression of Bax protein in bone marrow cells were significantly lower in the TDER group and granulocyte stimulating factor group(all P<0.05),while the proportion of cells in S-phase,the proportion of cells in G_(2)/M-phase and the expression of Bcl-2 protein in bone marrow cells were significantly higher in the TDER group and granulocyte stimulating factor group(all P<0.05);compared with the granulocyte stimulating factor group,the proportion of cells in S-phase and the expression of Bcl-2 protein in bone marrow cells were significantly lower in the TDER group(all P<0.05).Conclusion Tortoise-plastron decoction for enhancing the root can inhibit the apoptosis of bone marrow cells and induce them to enter the S-phase and G_(2)/M proliferation cycle,and promote bone marrow cell proliferation via up-regulating the expression of Bcl-2 protein and down-regulating the expression of Bax protein in bone marrow cells,thus to improve myelosuppression.
