应用SSR分子标记分析烟台地区黄瓜种质资源遗传多样性 被引量:3

Genetic diversity of cucumber germplasm resources in Yantai region was analyzed by SSR molecular markers

摘要 以22份黄瓜为材料,利用SSR分子标记技术进行遗传分析,构建22份黄瓜种质的指纹图谱和分子身份证。利用凝胶电泳技术,共筛选出9对SSR引物进行PCR扩增。9对引物一共检测到40个等位基因,每对引物检测出2~9个等位基因,多态性信息含量(PIC)平均为0.42。22份黄瓜种质的遗传相似系数范围是0.25~1.00,在相关系数为0.79时可将22份黄瓜种质分成六类,利用UPGMA法构建聚类分析图,所得分析结果与指纹图谱相符合。研究结果可进一步完善黄瓜种质资源收集与保存,为烟台地区黄瓜主栽品种鉴定和育种研究提供重要参考。 Using 22 cucumbers as materials,genetic analysis was carried out by using SSR molecular labeling technology to construct fingerprints and molecular ID cards of 22 cucumber germplasm resources.Using gel electrophoresis technology,a total of 9 pairs of SSR primer were screened for PCR amplification.A total of 40 alleles were detected in 9 pairs of primes,and 2 to 9 alleles were detected in each pair of primes,with an average PIC value of 0.42.The genetic similarity coefficient of 22 cucumber varieties ranges from 0.25 to 1.00.When the correlation coefficient is 0.79,22 cucumber germplasm can be divided into six categories.The UPGMA method is used to construct a cluster analysis map,and the obtained analysis results are consistent with the fingerprint.This study can further improve the collection and preservation of cucumber germplasm resources,and provide an important reference for the identification and breeding of cucumber varieties in Yantai.
