摘要
目的建立萹蓄HPLC-电化学检测(ECD)指纹图谱,并评价其抗氧化活性。方法萹蓄80%甲醇提取液的分析采用XBridge BEH shield RP18色谱柱(3.0 mm×150 mm,2.5μm);流动相乙腈-9.6 mg/mL柠檬酸溶液(pH 2.74),梯度洗脱;体积流量0.6 mL/min;柱温45℃;检测电压650 mV。采用铁离子还原能力、DPPH自由基清除能力和ABTS自由基清除能力评价抗氧化活性。再对指纹图谱共有峰与抗氧化活性进行灰色关联分析、偏最小二乘回归分析。结果17批样品指纹图谱中共有26个共有峰,通过与对照品比对指认出18种化学成分,相似度在0.944~0.991之间。表儿茶素、原花青素B2、原儿茶醛、儿茶素、洋蓟素、对香豆酸、异荭草苷、杨梅苷、槲皮苷、萹蓄苷等化学成分对抗氧化活性的贡献较大。结论HPLC-ECD指纹图谱结合谱效关系研究能明确萹蓄中不同化合物对抗氧化活性的贡献程度,可为萹蓄的质量控制提供参考依据。
AIM To establish the HPLC-ECD fingerprints for Polygonum aviculare L.,and to evaluate the antioxidant acitivity.METHODS The analysis of P.aviculare 80%methanol extract was performed on a 45℃thermostastic XBridge BEH shield RP18 column(3.0 mm×150 mm,2.5μm),with the mobile phase comprising of acetonitrile-9.6 mg/mL citric acid solution(pH 2.74)flowing at 0.6 mL/min in a gradient elution manner,and the detection voltage was set at 650 mV.The antioxidant activity was evaluated by ferric ion reducing antioxidant power(FRAP),DPPH radical scavenging capacity and ABTS radical scavenging capacity.Grey relational analysis(GRA)and partial least squares regression(PLSR)analysis were adopted in the common peaks of fingerprints and the antioxidant activity.RESULTS There were twenty-six common peaks in the fingerprints of the 17 batches of P.aviculare,and eighteen components were identified by comparison with the reference substance.The similarity ranged from 0.944 to 0.991.The components contributing greater to the antioxidant activity were epicatechin,proanthocyanidin B2,protoanthocatechualdehyde,catechin,cynarin,p-coumaric acid,isoorientin,myricetrin,quercitrin and avicularin,etc.CONCLUSION HPLC-ECD combined with the spectrum-effect method can provide a reference basis for the quality control of P.aviculare.
