摘要
利用计算机模拟技术筛选出牛乳的β-酪蛋白(β-casein)和羊乳的αS1-酪蛋白(α-S1-casein)中生物活性较好的多肽,并测定其抗氧化活性。先后运用Uniprot Database、BIOPEP-UWM Database、Peptide Ranker网络平台对多肽的潜在生物活性进行预测,并从中筛选出活性Score>0.5的多肽进行抗氧化活性测定。最终筛选出IPPL、VPPF、PPT、PPQ、PSF 5条多肽进行合成,并测定多肽的总抗氧化活性和·OH自由基清除能力测定。结果表明,5条多肽均具有一定的总抗氧化活性,其中2 g/L的PSF溶液的总抗氧化活性最强,相当于0.2085 mmol/L FeSO4。此外1 g/L的VPPF溶液的·OH清除率最高,为59.88%,该浓度下各多肽·OH清除能力排序为:VPPF(59.88%)>IPPL(46.97%)>PPQ(40.12%)>PSF(39.04%)>PPT(32.19%)。综上5条乳类多肽均具有一定的抗氧化活性,可作为抗氧化产品开发的研究对象;本文把计算机模拟与化学实验相结合的研究思路对多肽类功能性食品和药品的研发有着重要意义。
Oligopeptides fromβ-casein of bovine milk andα-S1-casein of goat milk with better biological activity protential were screened out by computer simulation technology,and their antioxidant activity were determined in vitro.After the the sequence files ofβ-casein from bovine milk andα-S1-casein from goat milk were obtained by Uniprot Database,GIOPEP-UWM Database was used for peptide simulated hydrolysis,then oligopeptides with 3-6 amino acids were screened to do biological activities prediction through Peptide Ranker network platform.Finally,five oligopeptides,IPPL,VPPF,PPT,PPQ and PSFS,whose score>0.5 have been selected for antioxidant activity determination.The results showed that a certain total antioxidant activity exhibited in five oligopeptides.In the maximum concentration of 2 mg/mL,the strongest effect was observed in PSF,which exhibited 0.2085 mmol/L FeSO4 relative concentration.In addition,the highest·OH scavenging ability(59.88%)was observed in the VPPF in the concentration of 1 mg/mL,in which·OH scavenging ability of other sequences can be listed as follows:IPPL(46.97%),PPQ(40.12%),PSF(39.04%),PPT(32.19%).In conclusion,five oligopeptides with certain antioxidant activity were observed,which may provide more possibilities for new antioxidant products research and development,which brings very broad value and prospects for functional food.
