Multi-index Integrated Evaluation Method Optimization of Percolation and ExtractionProcess of Bulleyacinitine A from Aconitum bulleyanumm

摘要 目的:多指标综合评分法优化直缘乌头中草乌甲素渗漉提取工艺。方法:在单因素试验基础上,以乙醇体积分数、浸泡时间、乙醇用量、渗漉流速为影响因素,草乌甲素转移率和干膏率为综合评价指标,正交试验优化提取工艺。采用HPLC测定草乌甲素含量,检测波长260 nm,流动相为0.2%三乙胺水溶液[用磷酸调节pH值(3.1±0.1)]-乙腈(60∶40)。结果:最佳条件为药材粉碎过10目筛,加3倍药材量70%乙醇浸泡时间16 h, 7倍药材量70%乙醇,渗漉流速2 mL/min,草乌甲素转移率>95%。结论:该方法简便稳定、重复性好,为草乌甲素的进一步分离纯化与利用提供实验依据。 Objective Multi-index Integrated Evaluation MethodOptimization of Percolation and Extraction Process of Bulleyacinitine A from Aconitum bulleyanumm.Methods Based on the analysis of the single factor experiment results,orthogonal test were used to determine the effects of ethanol dosage,ethanol concentration,soaking time and percolation speed on the extraction process of Aconitum bulleyanumm.by taking the dry extract yield and transfer rate of Bulleyacinitine A are taken as theindices for the comprehensive evaluation.HPLC was employed to determine contents of Bulleyacinitine A with detection wavelengths of 260 nm and mobile phases of was 0.2%triethylamine aqueous solution[pH(3.1±0.1)adjusted with phosphoric acid]-acetonitrile(60∶40).Results The optimum percolation process was to add 10 times amount of Aconitum bulleyanumm(sieve No.1)and the 70%of ethanol,and soak for 16 hours.The percolation speed was 2 mL/min per minute.Transferring rate of Bulleyacinitine A was more than 95%.Conclusion This optimized percolation process is simple and stable with good repeatability,it can provide a theoretical basis for Separation and purification of Bulleyacinitine A.
