清化肠饮对溃疡性结肠炎大鼠NLRP3/Caspase-1细胞焦亡通路的影响 被引量:4

Effect of Qinghuachang Yin(清化肠饮) on Nlrp3/Caspase-1 Cell Scorching Pathway in Ulcerative Colitis Rats

摘要 目的 探讨清化肠饮对溃疡性结肠炎模型大鼠NLRP3/Caspase-1细胞焦亡通路相关分子的影响。方法 将SPF级Wistar大鼠分为空白组和造模组,造模组采用“高脂高糖饮食+免疫”法构建湿热证UC大鼠模型,共持续20 d。造模成功后,造模组随机分为模型组、清化肠饮组、SASP组,每组8只。各组给予相应处理21 d后,麻醉采血,取结肠组织后,分别运用ELISA、qReal-time PCR法检测血清中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-18(IL-18)的含量及肠黏膜组织NLRP3、Caspase-1、ASC、GSDMD mRNA表达水平。结果 与空白组比较,模型组血清中IL-1β、IL-6、IL-18的含量及NLRP3、Caspase-1、ASC、GSDMD mRNA表达水平均显著升高(P<0.01);与模型组相比,清化肠饮组血清中炎症因子IL-1β、IL-6、IL-18表达水平及NLRP3、Caspase-1、ASC、GSDMD mRNA表达水平均显著降低(P<0.01)。结论 清化肠饮可通过抑制NLRP3/Caspase-1信号通路,阻断该信号通路相关分子的合成、激活,从而抑制UC大鼠肠道黏膜细胞焦亡程序,改善UC大鼠肠道黏膜损伤。 Objective To investigate the effect of Qinghuachang Yin(清化肠饮) on NLRP3/Caspase-1 cell scorching pathway related molecules in rats with ulcerative colitis model.Methods SPF-grade Wistar rats were divided into a blank group and a modeling group.The modeling group was constructed by the“high-fat,high-sugar diet+immunization”method,and lasted for 20 days.After successful modeling,the modeling group was randomLy divided into model group,Qinghuachang Yin group and SASP group,each group had 8 rats.After each group was treated accordingly for 21 days,blood was collected under anesthesia and colonic tissues were taken,and the levels of IL-1β,IL-6 and IL-18 in serum and the expression levels of NLRP3,Caspase-1,ASC and GSDMD mRNA in intestinal mucosa tissues were detected by ELISA and qReal-timePCR,respectively.Result Compared with the blank group,the levels of IL-1β,IL-6,IL-18 in serum and the expression levels of NLRP3,Caspase-1,ASC,GSDMDmRNA were significantly higher in the model group(P<0.01),compared with the model group,the expression levels of inflammatory factors IL-1β,IL-6,IL-18 in serum and the expression levels of NLRP3,Caspase-1,ASC,and GSDMD mRNA expression levels were significantly reduced in the Qinghuachang Yin group(P<0.01).Conclusion By inhibiting the NLRP3/Caspase-1 signaling pathway and blocking the synthesis and activation of molecules related to this signaling pathway,Qinghuachang Yin can inhibit the intestinal mucosal cell scorching program and improve the intestinal mucosal damage in UC rats.
