摘要
为研究油樟茎中内生真菌的群落组成及多样性,揭示不同季节对内生真菌群落组成和分布的影响,并探究菌株具有的功能,采用传统分离方法对4季油樟茎内生真菌进行分离、纯化及鉴定,并对所分离菌株群落组成及多样性进行统计分析,构建系统发育树,同时利用显色法和分光光度计法筛选产黄酮菌株,对功能较强菌株展开多酚含量测定、抗氧化能力、产功能酶和抑菌功能分析.结果表明:4季油樟茎部共分离得到98株内生真菌,隶属于2门3纲8目21科30属,Ascomycota为优势门,Alternaria为优势属,其中春季27株内生真菌隶属于1门13属,夏季20株内生真菌隶属于1门8属,秋季28株内生真菌隶属于1门13属,冬季23株内生真菌隶属于2门14属,4个季节共有Alternaria和Colletotrichum两属.多样性分析显示,冬季油樟茎内生真菌群落的多样性相对最丰富,夏季最低.对产黄酮功能筛选发现,15株具有产黄酮能力,其中Q73菌株产黄酮能力最强,且Q73表现出较好的抗氧化活性,具有产果胶酶、抑制草芽孢杆菌和抑制大肠杆菌的能力,Q73菌丝体黄酮和多酚含量均多于发酵液中的含量.因此,油樟茎部不同季节内生真菌存在共性和差异,为进一步挖掘油樟与其内生真菌之间的互作关系提供参考依据,且分离的菌株可用于后续次生代谢产物中化学成分的研究.
To study the community composition and diversity of endophytic fungi in Cinnamomum longepaniculatum stems,reveal the effects of different seasons on the community composition and distribution of endophytic fungi,and explore the functions of the strains,the endophytic fungi were isolated,purified and identified by the traditional isolation method.The community com-position and diversity of the isolated strains were analyzed statistically and phylogenetic trees were constructed.Meanwhile,the fla-vonoids producing strains were screened by chromogenic method and spectrophotometer,and the polyphenol content,antioxi-dant capacity,functional enzymes and antibacterial function of the functional strains were analyzed.The results show that 98 endo-phytic fungi are isolated from the stems of Cinnamomum longepaniculatum in 4 seasons,belonging to 2 phyla,3 classes,8 orders,22 families and 30 genera.Ascomycota is the dominant phyla,and Alternaria is the dominant genus.27 endophytic fungi belonged to 1 phyla,13 genera in spring,20 endophytic fungi belonged to 1 phyla,8 genera in summer,and 28 fungal strains belonged to 1 phylum and 13 genera in autumn,while 23 endophytic fungi belonged to 2 phylum and 14 genera in winter.Alternaria and Colletotrichum are present in all four seasons.Diversity analysis show that the diversity of endophytic fungi community is the high-est in winter and the lowest in summer.The screening of flavonoids production function show that 15 strains had the ability to produce flavonoids,among which strain Q73 had the strongest ability to produce flavonoids,and Q73 show better antioxidant ac-tivity,with the ability to produce pectinase,inhibit Bacillus cystoides and Escherichia coli.The contents of flavonoids and poly-phenols in mycelium Q73 are higher than those in fermentation broth.Therefore,there are similarities and differences between endophytic fungi in different seasons in the Cinnamomum longepaniculatum stems,which provide reference for further exploration of the interaction between Cinnamomum longepaniculatum and endophytic fungi,and the isolated strains could be used for the subse-quent study of chemical constituents in secondary metabolites.
